摘  要:为研究不同清洗处理对荠菜中细菌微生物多样性的影响,采用Illumina高通量测序技术研究荠菜样本中细菌群落结构,结合菌落总数分析不同清洗处理对荠菜的减菌效果。次氯酸钠、酸性电解水和臭氧水清洗能够显著降低荠菜菌落总数,减菌率分别为95.43%、98.00%、99.06%。对不同清洗处理荠菜样品的16S rDNA V3~V4区域进行测序,共获得13个门、176个属。在“属”水平上,Brassica在荠菜细菌群落结构中占据绝对核心优势,Boechera次之;次氯酸钠处理后Brassica丰度显著降低,从88.03%降至84.21%;酸性电解水清洗后Brassica丰度略微上升,为89.22%;而臭氧水对Brassica丰度无显著影响。在“门”水平上,Streptophyta是荠菜细菌群落结构核心菌门,次氯酸钠处理后Streptophyta丰度降低,从94.11%降至89.80%;酸性电解水、次氯酸钠、臭氧水处理均降低变形菌门丰度,从3.38%分别降低到2.78%、3.16%、2.39%。3种清洗处理对荠菜都具有较好的减菌效果,臭氧水清洗荠菜减菌率较高,而次氯酸钠处理能够降低核心菌属和菌门丰度,表明荠菜样品核心菌群对次氯酸钠耐受度低。综合分析得出,臭氧水和次氯酸钠是适宜清洗荠菜的加工方式。In order to observe the influence of different cleaning methods on the bacterial microbial diversity in Capsella bursa-pastoris,Illumina high-throughput sequencing technology was used to study the bacterial community structure,and to evaluate the safety of vegetables in different cleaning treatments based on the total number of bacterial colonies.Sodium hypochlorite,acid electrolytic water and ozone water cleaning significantly reduced the total number of Capsella bursa-pastoris bacterial colonies,the bacteria reduction rate was 95.43%,98.00%,and 99.06%,respectively.A total of 13 phyla and 176 genera were obtained by sequencing the 16S rDNA V3–V4 region of the different wash-treatment samples.At the classification level of“genus”,Brassica occupied the absolute core advantage in the bacterial community structure of Capsella bursa-pastoris,followed by Boechera,and the abundance of Brassica decreased significantly after sodium hypochlorite treatment,from 88.03%to 84.21%.After acid electrolytic water treatment,the Brassica abundance increased slightly to 89.22%,while ozone water had no significant effect on Brassica abundance.At the classification level of“phylum”,Streptophyta is the core phylum of Capsella bursa-pastoris bacterial community structure.Streptophyta abundance decreased from 94.11%to 89.80%,after sodium hypochlorite treatment.Acidic electrolytic water,sodium hypochlorite and ozone water treatment all reduced the abundance of Proteobacteria from 3.38%to 2.78%,3.16%and 2.39%respectively.Three cleaning methods for Capsella bursa-pastoris have a better bacteria reduction effect,ozone water cleaning had a higher reduction rate,while the sodium hypochlorite treatment reduced the abundance of core bacteria and phylum,indicating that the tolerance of the core bacteria group of Capsella bursa-pastoris sample to sodium hypochlorite is low.Comprehensive analysis shows that ozone water and sodium hypochlorite are suitable processing methods for cleaning Capsella bursa-pastoris.
