摘  要:以清香型大曲为研究对象,采用稀释涂布平板法从中分离酵母菌,对其形态特征进行观察,采用2,3,5-氯化三苯基四氮唑(TTC)显色法初筛,通过发酵力、产酒及产香能力分析进行复筛。对筛选酵母菌进行分子生物学鉴定及耐高温、耐乙醇、耐高糖、耐酸能力检测,筛选出耐受性优良的功能性酵母菌。结果表明,从清香型大曲中共分离初筛22株酵母菌,复筛后得到4株功能酵母菌,菌株B-6、C-3、D-10、D-11分别被鉴定为费比恩塞伯林德纳氏酵母(Cyberlindnera fabianii)、酿酒酵母(Saccharomyces cerevisiae)、扣囊复膜孢酵母(Saccharomycopsis fibuligera)、异常威克汉姆酵母(Wickerhamomyces anomalus)。耐受性分析结果表明,菌株B-6和C-3可耐受39℃、pH值为3、酒精度12%vol、葡萄糖质量浓度400 g/L环境条件。因此,菌株B-6和C-3可作为潜在功能菌株应用于白酒生产中。The yeasts were isolated from the light-flavor Daqu using the dilution-coated plate method,and their morphological characteristics were observed.Initially,the yeasts were screened using the 2,3,5-triphenyltetrazolium chloride(TTC)color development method,and rescreened by analysis of fermentation power,alcohol production and aroma production capacity.The screened yeasts were identified by molecular biology and tested for high temperature tolerance,ethanol tolerance,high sugar tolerance and acid tolerance,and the functional yeasts with good tolerance were screened.The results revealed the isolation of a total of 22 yeast strains from the light-flavor Daqu during the primary screening.After rescreening,four functional yeast strains were obtained,with strains B-6,C-3,D-10,and D-11 identified as Cyberlindnera fabianii,Saccharomyces cerevisiae,Saccharomycopsis fibuligera and Wickerhamomyces anomalus,respectively.Tolerance analysis demonstrated that strains B-6 and C-3 exhibited resilience to environmental conditions of temperature 39℃,pH 3,alcohol content 12%vol and glucose mass concentration 400 g/L.Therefore,strains B-6 and C-3 could be used as potential functional strains in Baijiu production.
