摘 要:利用超声波辅助酶法提取山香圆叶中的总黄酮,选取纤维素酶添加量、超声时间、酶解时间为提取因素,以黄酮提取率为响应值,在单因素实验的基础上,通过Box-Behnken响应面法设计三因素三水平的优化试验。结果表明:优化后的工艺为纤维素酶添加量4mg、超声时间43min、酶解时间63min,在此条件下测得山香圆叶黄酮提取率为2.65%。该方法提取工艺简便、高效、没有有机溶剂残留,可为山香圆叶中总黄酮的提取提供参考。The total flavonoids were extracted by ultrasonic assisted enzyme method.Cellulase content,ultrasonic time and enzymatic hydrolysis time were selected as the extraction factors,and the extraction rate of flavonoids was taken as the response value.On the basis of single factor experiment,a three-factor and three-level optimization experiment was designed by Box-Behnken response surface method.The results showed that the optimized process consisted of 4 mg cellulase,43 min ultrasonic time and 63 min enzymatic hydrolysis time.Under these conditions,the extraction yield of flavonoids was 2.65%.The extraction process is simple,efficient and free of organic solvent residue,which can provide a reference for the extraction of total flavonoids from the leaves of rotunda.
