摘  要:目的研究不同因素对菊花清除自由基能力的影响,鉴定其功能性成分,探索菊花对羟基自由基引导的2-脱氧核糖裂解的抑制作用。方法测定菊花提取液在不同浓度、温度和反应时间条件下对1,1-二苯基-2-苦基肼自由基(1,1-diphenyl-2-picrylhydrazyl radical,DPPH·)和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐阳离子自由基[2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)ammonium salt cationic radical,ABTS^(+)·]的清除率。采用高效液相色谱法,测定了菊花提取液与DPPH·和ABTS^(+)·反应前后供试品溶液中木犀草苷、木犀草素葡萄糖醛酸苷、绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C的含量。采用体外Fenton反应实验体系,测定菊花提取液对2-脱氧核糖裂解的保护作用。结果菊花样品浓度、温度和反应时间对DPPH·和ABTS^(+)·的清除率均有影响。与DPPH·和ABTS^(+)·反应后,菊花提取液中7种测定成分的含量均明显降低,是菊花清除自由基的主要功能性成分。菊花提取液对2-脱氧核糖裂解的保护作用与剂量相关。结论菊花具有清除自由基、抑制2-脱氧核糖裂解的作用,研究结果为菊花的深入开发提供了依据。Objective To study the influences of different conditions on free radical scavenging rates of Chrysanthemum morifolium,identify its functional components,and explore its inhibitory effects on 2-deoxyribose degradation mediated by hydroxyl radical.Methods Chrysanthemum morifolium extract scavenging rates of 1,1-diphenyl-2-picrylhydrazyl radical(DPPHꞏ)and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)ammonium salt cationic radical(ABTS^(+)ꞏ)were measured under different sample concentration,temperature and reaction time.High performance liquid chromatography was used to determine the content of luteolin,luteolin glucuronide,chlorogenic acid,cryptochlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C in Chrysanthemum morifolium extract solutions before and after the reaction with DPPHꞏand ABTS^(+)ꞏ.The protective effects of Chrysanthemum morifolium extract on 2-deoxyribose degradation was determined by Fenton reaction experiment system in vitro.Results Sample concentration of Chrysanthemum morifolium,reaction temperature and reaction time were positive correlated to the scavenging rates of DPPHꞏand ABTS^(+)ꞏ.After reacting with DPPHꞏand ABTS^(+)ꞏ,the content of 7 kinds of components in Chrysanthemum morifolium extract were obviously reduced,which was the main functional component of Chrysanthemum morifolium for scavenging free radicals.The inhibition of Chrysanthemum morifolium on 2-deoxyribose degradation was dose-dependent.Conclusion Chrysanthemum morifolium extract has activities of scavenging free radicals and inhibition on 2-deoxyribose degradation,which provides the basis for further development of Chrysanthemum morifolium.
