摘 要:目的 调查2021年广州市番禺区湿米粉和湿淀粉制品中椰毒假单胞酵米面亚种的污染情况,探讨基质辅助激光解析电离飞行时间质谱仪(matrix-assisted laser desorption ionization time of flight mass spectrometry, MALDI-TOF MS)法和实时荧光定量PCR法的应用价值。方法 按照《食品安全国家标准食品微生物学检验唐菖蒲伯克霍尔德菌(椰毒假单胞菌酵米面亚种)检验》(GB/T 4789.29-2020)和《食品安全国家标准食品中米酵菌酸的测定》(GB 5009.189-2016)的要求进行检验。采用全自动微生物鉴定系统VITEK2 COMPACT生化法和MALDI-TOF MS法鉴定样品中的唐菖蒲伯克霍尔德菌,用实时荧光定量PCR法检测椰毒假单胞菌产毒bon基因和高效液相色谱法检测米酵菌酸。结果 40份样品中有2份湿米粉和1份湿淀粉制品检出唐菖蒲伯克霍尔德菌,1份湿米粉样品检出椰毒假单胞酵米面亚种。在唐菖蒲伯克霍尔德菌的鉴定中,MALDI-TOF MS法和实时法与传统生理生化检测法结果一致。结论 番禺区湿米粉和湿淀粉制品存在被椰毒假单胞酵米面亚种污染的风险。MALDI-TOF MS法和实时荧光定量PCR方法在鉴定该菌时更快速、结果更准确。Objective To investigate the pollution of Pseudomonas cocovenenans subsp.farinofermentans in wet rice flour and wet starch products in Panyu District,Guangzhou in 2021,and to explore the application value of matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS)method and real-time fluorescence quantitative PCR method.Methods According to the requirements of National Standards for Food Safety Food Microbiology Tests Burkholderia gladiolus(Pseudomonas cocovenenans subsp.farinofermentans)Test(GB/T 4789.29-2020)and National Standards for Food Safety Determination of Bongkrekic Acid in Foods(GB 5009.189-2016),the identification of Burkholderia gladiolus in samples was conducted by VITEK2 COMPACT biochemical method and MALDI-TOF MS method.The real-time fluorescence quantitative PCR was used to detect the toxin-producing bon gene of Pseudomonas cocovenenans,and the high-performance liquid chromatography was used to detect the Bongkrekic acid.Results In the forty samples,Burkholderia gladiolus were detected in two wet rice flour samples and one wet starch product,and Pseudomonas cocovenenans subsp.farinofermentans was detected in one wet rice flour sample.In the identification of Burkholderia gladiolus(Pseudomonas cocovenenans subsp.farinofermentans),the results of the MALDI-T-OF MS and the real-time fluorescence quantitative PCR were consistent with traditional physiological and biochemical detection methods.Conclusion The wet rice flour and wet starch products in Panyu District are at risk of being contaminated by Pseudomonas cocovenenans subsp.farinofermentans.The MALDI-TOF MS method and the real-time fluorescence quantitative PCR method are more rapid and accurate in the identification of this bacteria.
