摘 要:以自然干燥山银花为原料,采用超声波辅助乙醇溶剂提取法,通过单因素试验,结合响应面对山银花多酚的提取工艺进行优化,并用超高效液相色谱串联质谱(UPLC-MS/MS)基于植物广泛靶向数据库对其多酚物质进行定性定量初鉴。结果表明,山银花总酚提取量的最佳工艺条件为:料(g)液(mL)比10∶1,乙醇浓度57%,超声功率70 W,提取温度60℃,提取时间11 min,此条件下山银花总酚提取量为71.08 mg/g;山银花酚酸以绿原酸同分异构体为主,黄酮以川陈皮素、木犀草苷和香叶木苷为主。The natural dry Lonicerae flos was used as raw material.Ultrasonic-assisted ethanol solvent extraction method was adopted to optimize the extraction process of polyphenols from Lonicerae flos through a single factor experiment combined with the response surface method,and UPLC-MS/MS was utilized to conduct a preliminary qualitative and quantitative identification of its phenolic substances based on the plant's extensive target database.The results show that the optimal process conditions for the extraction of total phenols from Lonicerae flos are as follows:solid-liquid ratio of 10 ∶ 1 g/mL,ethanol concentration of 57%,ultrasonic power of 70 W,extraction temperature at 60°C,and extraction time of 11 min.The amount of extracted total phenols from Lonicerae flos is 71.08 mg/g;the phenolic acid of Lonicerae flos is mainly chlorogenic acid isomers,and the flavonoids are mainly tangerine,luteolin,and diosmin.
