摘 要:以信阳水牛乳为原料,采用高速剪切分散均质法提取唾液酸。以唾液酸得率为指标,在单因素试验的基础上通过响应面法优化提取工艺,同时研究其对记忆障碍小鼠的改善作用。结果表明:最佳提取工艺条件为剪切转速14000 r/min、剪切时间100 s、剪切次数3次,在此条件下,水牛乳唾液酸得率为9.48%。小鼠记忆障碍改善作用试验结果表明,水牛乳唾液酸组小鼠的逃逸潜伏期明显减少,海马乙酰胆碱(Ach)含量明显升高,乙酰胆碱酯酶(AchE)活力明显降低,表明水牛乳唾液酸具有较好的改善记忆作用。Using Xinyang buffalo milk as raw material,sialic acid was extracted by high speed shear dispersion homogenization method.Taking the yield of sialic acid as an indicator,the extraction process was optimized by response surface methodology based on single factor experiments,and its improvement effect on memory impaired mice was studied.The results showed that the optimal extraction process conditions were shear speed 14000 r/min,shear time 100 s,and shear times 3.Under the conditions,the yield of sialic acid from buffalo milk was 9.48%.The experimental results of the improvement effect of water buffalo milk sialic acid on memory impairment in mice showed that the escape latency of the mice in the water buffalo milk sialic acid group was significantly reduced,the acetylcholine(Ach)content in the hippocampus was significantly increased,and the acetylcholinesterase(AchE)activity was significantly reduced,indicating that water buffalo milk sialic acid had a good effect on memory improvement.
