两种方法制备的蛹虫草小分子肽分子量分布及体外抗氧化能力对比研究

摘  要:目的探究复合酶解法与酶-高压蒸煮耦合水解法制备蛹虫草小分子肽的分子量分布情况与体外抗氧化活性差异。方法采用凝胶色谱法检测两组肽分子量分布情况,并分别测定总抗氧化能力(T-AOC),羟基自由基和超氧阴离子自由基清除能力,综合评价两组多肽体外抗氧化能力。结果两种方法制备的肽样品分子量分布情况相近,分子量<10×10^(3) Da的组分占比近60%,分子量<5×10^(3) Da的组分占比约31%。两种方法制得的分子量<10×10^(3) Da、分子量<5×10^(3) Da的蛹虫草活性肽的清除羟基自由基和超氧阴离子自由基能力均优于还原性谷胱甘肽(GSH)。其中复合酶解法所得分子量<5×10^(3)活性多肽的T-AOC活性明显高于GSH。结论两种方法制备的多肽分子量分布无明显差异,且均具有良好的体外抗氧化活性。Objective To investigate the molecular weight distribution of small molecular peptides from Cordyceps militaris prepared by compound enzymatic hydrolysis and enzyme-high pressure steaming coupled hydrolysis and the difference of antioxidant activity in vitro.Methods The molecular weight distribution of peptides obtained by two methods was detected by gel chromatography,and the total antioxidant capacity(T-AOC),hydroxyl radical and superoxide anion radical scavenging capacity were measured respectively to evaluate the in vitro antioxidant capacity of the two groups of peptides.Results The molecular weight distribution of peptide samples prepared by two methods was similar.The proportion of components with molecular weight less than 10×10^(3) Da was near 60%,and the proportion of components with molecular weight less than 5×10^(3) Da was about 31%.The active peptides with molecular weight less than 10×10^(3) Da and molecular weight less than 5×10^(3) Da prepared by two methods showed better hydroxyl radical and superoxide anion radical scavenging ability than reducing glutathione(GSH).The T-AOC activity of active peptides with molecular weight less than 5×10^(3) Da obtained by compound enzymatic hydrolysis was significantly higher than that of GSH.Conclusion The molecular weight distribution of peptides prepared by the two methods showed no significant difference,and both had good in vitro antioxidant activity.
