摘 要:构建了以组氨酸(H)-色氨酸(W)-组氨酸(H)-组氨酸(H)-谷氨酸(E)为氨基酸序列的荧光多肽探针HWHHE,结合酶标仪,可实现对水溶液中Cu^(2+)的快速高通量检测。结果表明:在优化的检测条件下(25℃、pH 7.4),探针多肽荧光HWHHE强度与Cu^(2+)浓度(0~1.0μmol/L)呈线性关系(R^(2)=0.997),检出限为2.67μg/L,远低于国家标准规定的限量,且加标回收率为93.7%~102.1%。采用高通量法检测实际样品(自来水以及饮用水)中Cu^(2+)浓度,其结果与用分光光度法测定结果无显著差异(P>0.05),证明荧光多肽探针HWHHE可结合酶标仪实现水溶液中Cu^(2+)的高通量快速检测。A fluorescent polypeptide probe HWHHE was constructed with an amino acid sequence of histidine(H)-tryptophan(W)-histidine(H)-histidine(H)-glutamic acid(E).Combined with microplate reader,it could achieve rapid and high-throughput detection of Cu^(2+)in aqueous solutions.The results showed that under optimized detection conditions(25℃,pH 7.4),the fluorescence intensity of HWHHE probe had a linear relationship with the Cu^(2+)concentration(0-1.0μmol/L)(R^(2)=0.997).The limit of detection was 2.67μg/L,which was lower than the allowable limit of national standard,and the recovery rate of the HWHHE fluorescence polypeptide probe for the determination of Cu^(2+)was 93.7%-102.1%.The high-throughput method was used to detect the content of Cu^(2+)in actual samples(tap water and drinking water),and the results had no significant difference with the results detected with spectrophotometric measurement(P>0.05),which suggested that the HWHHE fluorescent polypeptide probe could be combined with a microplate reader to achieve rapid and high-throughput detection of Cu^(2+)in aqueous solutions.
