摘  要:曲酸是一种真菌次生代谢产物,在化妆品、食品和制药工业中有多种用途。为揭示米曲霉发酵生产曲酸的生物合成机制,该研究利用基因工程的手段获得一株kojR过表达菌株T58号,摇瓶发酵曲酸产量最高达到29.54 g/L,较原始米曲霉菌株3.042的10.12 g/L提升191.9%。将发酵过程中获得的菌株T58与3.042菌丝体进行转录组分析,发现两者之间存在2450个显著差异表达基因,其中1203个基因表达上调,1247个基因表达下调。菌株T58差异性表达量最高的前20个基因中,上调基因主要为不同类别的氧化还原酶,与氮源利用相关基因的转录水平显著下调。基因本体数据库分析显示,与转运活性相关的差异基因数量最多。Kojic acid,a fungal secondary metabolite,has wide applications in the cosmetic,food,and phar⁃maceutical industries.To elucidate the biosynthesis mechanism of kojic acid in Aspergillus oryzae through fer⁃mentation,a kojR overexpressed strain named T58 was obtained using genetic engineering.During shaking flask fermentation,T58 achieved a peak kojic acid yield of 29.54 g/L,increasing by 191.9%compared with the original A.oryzae strain 3.042,which produced only 10.12 g/L.Transcriptome analysis on both strain T58 and 3.042 mycelium obtained during fermentation revealed 2450 significantly differentially expressed genes in T58 compared with the control.Among them,1203 genes were up⁃regulated while 1247 genes were down⁃regulated.Among the top 20 genes with the most pronounced expression in strain T58,the up⁃regulated genes were mainly different types of oxidoreductases,and the transcript levels of genes related to nitrogen source uti⁃lization were significantly down⁃regulated.Gene ontology analysis highlighted a substantial number of differen⁃tially expressed genes related to transporter activity.
