摘 要:以陕西茯砖黑毛茶为原料,基于溶剂萃取法的单因素实验对有效成分进行提取,通过正交实验测定陕西茯茶提取物对DPPH和ABTS+自由基的清除率来反映其有效成分的抗氧化能力。研究结果表明利用乙醇-水溶液萃取法获得的提取物具有较强的抗氧化成分,抗氧化成分提取的最佳条件为:40%的乙醇溶液,提取温度45℃、提取时间45 min、料液比为1∶60。研究发现该有效成分对DPPH的清除率高达85%,高于对ABTS+自由基的清除率(约67%)。对茯茶素分子的化学物理属性与抗氧化的关联性进行了定量构效关系的研究,揭示出含有酚羟基的茯茶素分子的抗氧化性与分子中氢键给体的数量存在线性关系。总之,本文对陕西茯茶中多酚的提取工艺以及抗氧化性能的定量构效关系(QSAR)的研究可以为陕西茯茶抗氧化产品的开发提供理论依据。Using Shaanxi Fu brick tea as the raw material,the active ingredients were extracted based on a solvent extraction method through single-factor experiments.The antioxidant capacity of the effective components was reflected by the clearance rate of DPPH and ABTS+free radicals in Shaanxi Fu tea extract,determined through orthogonal experiments.The findings revealed that the extract obtained using an ethanol-water solution extraction method possessed high antioxidant capacity.The optimal conditions for extracting antioxidant components are as follows:a 40% ethanol solution,extraction time of 45℃,extraction time of 45 minutes,and a solid-to-liquid ratio of 1∶60.The extract showed an 85%clearance rate for DPPH radicals,which was higher than the clearance rate of 67% for ABTS+radicals.A quantitative structure-activity relationship study was conducted on the chemical and physical properties of the catechin molecules to elucidate the correlation between these properties and antioxidant activity,revealing a linear relationship between the antioxidant properties of catechin molecules containing phenolic hydroxyl groups and the number of hydrogen bond acceptors in the molecule.This research on the extraction and QSAR of antioxidant components in Shaanxi Fu tea lays a theoretical foundation for developing antioxidant products from this tea.
