摘 要:采用凯氏定氮法检测杏鲍菇中蛋白质含量,并通过单因素和响应面试验对消化时间、消化温度及催化剂用量等条件进行了优化。结果显示,凯氏定氮法检测杏鲍菇中蛋白质含量的最优条件参数为消化时间1.85 h,消化温度438℃,催化剂浓硫酸用量14.8 mL,该条件下测定的杏鲍菇中蛋白质含量最高,为20.71 g/100 g。与国标法比较,本研究中优化后的方法提高了蛋白质含量的检测效率,减少了催化剂浓硫酸的用量,缩短了消化时间。The Kjeldahl nitrogen determination method was used to detect the protein content of Pleurotus eryngii,and the digestion time,digestion temperature and catalyst dosage were optimized by single factor and response surface tests.The results showed that the optimal parameters for the detection of protein content in Pleurotus eryngii by Kjeldahl nitrogen determination method were the digestion time of 1.85 h,the digestion temperature of 438℃,and the amount of concentrated sulfuric acid in the catalyst was 14.8 mL,and the protein content of Pleurotus eryngii was the highest(20.71 g/100 g)under these conditions.Compared with the national standard method,the optimized method in this study improved the detection efficiency of protein content,reduced the amount of concentrated sulfuric acid in the catalyst,and shortened the digestion time.
