摘  要:采用愈创木酚固体培养基对分离自栽培食用菌菌棒污染菌的18株木霉菌所产木质素降解酶活性进行了研究,筛选出具有显著木质素降解酶活性的木霉菌株Z-1。通过形态特征观察和分子鉴定,确定该菌株为深绿木霉Trichoderma atroviride。通过对该菌株液体发酵产酶活性的研究发现,以产漆酶液体培养基为基础培养基,在250 mL锥形瓶50 mL装样量的条件下,其产酶活性最佳培养条件为温度20℃、摇床转速90 r/min、接菌量2%(体积比)、pH 6、时间6 d。本研究为进一步开展高产木质素降解酶木霉菌株的应用研究提供了理论基础。Guaiacol solid culture medium was applied to study the lignin-degrading enzyme characteristics of 18 strains of Trichoderma strains isolated from the contaminated synthetic rods cultivating edible mushrooms,and a strain Trichoderma Z-1 with significant lignin-degrading activity was found.Based on morphological characteristics and rDNA sequence analysis,it was identified as Trichoderma atroviride.Liquid fermentation experiments found that the optimal culture conditions of the strain for enzyme production were temperature of 20℃,shaking speed of 90 r/min,inoculation rate of 2%,and fermenting time of 6 days under pH 6 in a 250 mL conical flask containing 50 mL of laccase producing liquid culture medium.Further application study of the lignin-degrading enzyme of Trichoderma atroviride Z-1 is in progress.
