摘 要:研究大孔树脂对山药皮多酚的纯化工艺,并评价纯化后的多酚对DPPH·、ABTS^(+)·、·O^(-)_2、·OH的清除能力。结果表明:X-5为纯化山药皮多酚的最佳树脂;最佳的纯化工艺流程为上样液pH 6.0、上样液质量浓度0.6 mg/mL、乙醇(洗脱液)体积分数80%、上样流速2 mL/min、洗脱流速2 mL/min;其吸附等温线符合Langmuir单分子层吸附;经此条件纯化后,其多酚纯度从34.09%提高到80.22%。抗氧化活性试验表明,山药皮多酚纯化物对DPPH·、ABTS^(+)·、·OH和·O^(-)_2清除能力均明显高于粗提物,具有较强的抗氧化活性,是具有开发潜力的天然抗氧化剂。The purification process of yam peel polyphenol using macroporous resin was studied,and the scavenging ability of purified polyphenol to DPPH·,ABTS^(+)·,·O^(-)_(2)and·OH was evaluated.The results showed that X-5 was the best resin for purifying yam peel polyphenol.The optimal purification process was pH 6.0 of sample solution,mass concentration 0.6 mg/mL of sample solution,volume fraction of ethanol(elution)80%,sample flow rate 2 mL/min,elution flow rate 2 mL/min.The adsorption isotherm was consistent with Langmuir monolayer adsorption.After purification under these conditions,the purity of polyphenols extract was increased from 34.09%to 80.22%.The antioxidant activity test showed that the purified yam peel polyphenols had significantly higher scavenging capacity for DPPH·,ABTS^(+)·,·OH and·O^(-)_(2)than the crude extract,and had strong antioxidant activity,which was a natural antioxidant with potential development.
