摘  要:以宣木瓜为原料,采用超声波辅助提取宣木瓜中超氧化物歧化酶(superoxide dismutase,SOD),以pH、料液比、浸提时间和超声时间为试验因素,以SOD酶活力为试验指标,采用四因素三水平正交试验,优化宣木瓜SOD的提取工艺参数;对宣木瓜SOD抗氧化活性进行初步研究,测定·OH清除能力,并探讨其对过氧化氢(H_(2)O_(2))诱导的原代小鼠空肠黏膜上皮细胞氧化应激的保护作用,将细胞分为H_(2)O_(2)+SOD组(60 mg/L的SOD预处理24 h,3%H_(2)O_(2)处理24 h)和H_(2)O_(2)组(3%H_(2)O_(2)处理24 h),采用Calcein AM/PI染色法检测细胞活性。结果表明:宣木瓜SOD的提取最佳条件为磷酸盐缓冲液p H 6.8、料液比1∶6(g/mL)、浸提时间2 h、超声时间40 min,此时宣木瓜中SOD酶活力为289.28(U/g),·OH清除能力最高达到97.26%,且宣木瓜SOD对H_(2)O_(2)诱导的原代小鼠空肠黏膜细胞氧化应激损伤具有明显保护作用。Selection of Chaenomeles speciosa as raw material,ultrasound was used to assist in the extraction of superoxide dismutase(SOD)from Chaenomeles speciosa.Orthogonal experiment of four factors and three levels was used to optimize the extraction process parameters of SOD in Chaenomeles speciosa,with the pH,solid-liquid ratio,extraction time and ultrasonication time as test factors and SOD viability as experimental indexes.Meanwhile,we initially investigated the antioxidant activity of SOD in Chaenomeles speciosa,including scavenging activity on OH radical and the protective effect of SOD on the oxidative damage in primary mouse jejunal mucosal epithelial cells induced by H_(2)O_(2).The cells were divided into the H_(2)O_(2)+SOD group(60 mg/mL SOD pretreatment for 24 h,3%H_(2)O_(2)treatment for 24 h)and the H_(2)O_(2)group(3%H_(2)O_(2)treatment for 24 h).The cell activity was detected by Calcein AM/PI staining.The results showed that the best extraction conditions were phosphate buffer pH 6.8,solid-liquid ratio 1∶6(g/mL),extraction time 2 hours,and ultrasound time 40 minutes,the enzyme activity of SOD in Chaenomeles speciosa was 289.28(U/g),and SOD in Chaenomeles speciosa had strong hydroxyl radical scavenging(97.26%,1 mg/mL)and an obvious protective effect on H_(2)O_(2)-induced oxidative stress damage in primary mouse jejunal mucosal epithelial cells.
