摘  要:类黄酮物质是茶树最重要的次生代谢物,对茶叶风味和保健功能具有决定作用。尿苷二磷酸糖基转移酶(uridine diphosphate glycosyltransferase,UGT)催化的糖基化反应是黄酮类物质合成的关键步骤,具有重要的研究价值。本研究应用Iso-seq全长转录组技术获得了茶树UGT基因,通过与拟南芥以及已经报道的合成类黄酮的UGT基因进行序列比对,筛选参与类黄酮生物合成的UGT基因,并采用RT-qPCR和高效液相色谱测定候选基因的表达和类黄酮物质的含量,分析基因表达量与类黄酮物质含量的相关性。结果发现UDP75L121、UDP75L123、UDP78A15、UDP94P11与儿茶素没食子酸酯(catechin gallate,CG)和表儿茶素没食子酸酯(epicatechin gallate,ECG)含量呈正相关,UDP78A15与花青素矢车菊色素(cyanidin,Cy)、天竺葵色素(pelargonidin,Pel)和锦葵色素(malvidin,Ma)含量呈正相关,而UDP75L123、UDP78A15与黄烷醇类山柰酚(kaempferol,Kae)和酚酸类鞣花酸(ellagic acid,EA)含量呈负相关(P<0.05),初步表明这4个基因可能参与茶叶类黄酮物质的生物合成。本研究可为进一步分析茶树类黄酮物质的生物合成提供参考。Flavonoids are the most important secondary metabolites in Camellia sinensis,which are responsible for the flavor and healthful functions of tea.Uridine diphosphate glycosyltransferase(UGT)catalyzes the glycosylation of flavonoids,which is a key step in flavonoid synthesis.Therefore,UGT-genes regulate the contents of flavonoids,and have important research values.In this work,genes encoding UGT-glycosyltransferases in Camellia sinensis were obtained through Iso-seq full-length transcriptome analysis,and UGT genes involved in flavonoid biosynthesis were identified by sequence alignment of UGT genes in Arabidopsis thaliana and the known genes involved in synthesis of flavonoids.The expressions of candidated genes and the contents of flavonoids were also detected by RT-qPCR and high performance liquid chromatography,then the correlation between gene expressions and the contents of flavonoids were further analyzed.The results revealed that expression of UDP75L121,UDP-75L123,UDP78A15,and UDP94P11 were positively correlated with the contents of catechin gallate(CG)and epicatechin gallate(ECG);and expression of UDP78A15 was positively correlated with the contents of anthocyanins cyanidin(Cy),pelargonidin(Pel)and malvidin(Ma),while UDP75L123 and UDP78A15 were negatively correlated with the flavanol kaempferol(Kae)and phenolic acid ellagic acid(EA)contents(P<0.05).The results indicat that these four genes might be involved in the biosynthesis of flavonoids in Camellia sinensis,which provided a basis for further study the biosynthesis of flavonoids in Camellia sinensis.
