摘  要:为了优化刺五加多糖的复合酶提取工艺,以刺五加多糖得率作为指标,采用正交实验确定复合酶添加量,并基于响应面法对复合酶提取刺五加多糖的工艺进行优化。通过刺五加多糖对α-葡萄糖苷酶的抑制情况考察其降糖活性。结果显示复合酶(木瓜蛋白酶240 mg、果胶酶170 mg、纤维素酶160 mg)提取刺五加多糖的最佳工艺条件为酶解时间2.1 h、酶解pH 6.0、料液比1∶53 g/mL、酶解温度49℃。在此工艺下刺五加多糖得率为14.21%±0.09%,与预测值14.259%接近。提取条件各因素之间的主次顺序为料液比>酶解温度>酶解时间>酶解pH。体外活性实验表明,在0.1~0.5 mg/mL范围内,刺五加多糖对α-葡萄糖苷酶抑制率随着浓度的增大呈上升趋势,最大可达到65.83%±0.08%,IC_(50)为0.177 mg/mL,接近于阿卡波糖。由此可知复合酶提取刺五加多糖工艺稳定、可靠,刺五加多糖对α-葡萄糖苷酶具有一定抑制作用。In order to optimize the compound enzymes extraction process of Acanthopanax senticosus polysaccharides(ASPs),with the yield of ASPs as the index,and the orthogonal experiment was used to determine the amount of compound enzyme added,and the process of extracting ASPs by compound enzymes was optimized based on the response surface method.The inhibitory activity of ASPs onα-glucosidase was investigated to assess its hypoglycemic activity.The results showed that the optimal conditions for the extraction of ASPs with compound enzymes(papain 240 mg,pectinase 170 mg,cellulase 160 mg)were as follows:enzymatic hydrolysis time of 2.1 h,enzymatic hydrolysis pH of 6.0,solid-to-liquid ratio of 1∶53 g/mL,and enzymatic hydrolysis temperature of 49℃.Under this process,the yield of ASPs was 14.21%±0.09%,which was close to the predicted value of 14.259%.The primary and secondary order among the extraction conditions was the ratio of solid-to-liquid>enzymatic hydrolysis temperature>enzymatic hydrolysis time>enzymatic hydrolysis pH.In vitro activity experiments showed that within the range of 0.1〜0.5 mg/mL,the inhibition rate of ASPs onα-glucosidase increased with the increase of concentration,reaching a maximum of 65.83%±0.08%,and the IC_(50)was 0.177 mg/mL,which was close to that of acarbose.It can be concluded that the optimized complex enzyme extraction process for ASPs was stable and reliable.Furthermore,the extracted ASPs demonstrated a notable inhibitory effect onα-glucosidase.
