摘 要:以橘皮为原料,通过响应面优化了碱法提取橘皮不溶性膳食纤维的工艺,对膳食纤维结合胆固醇胶束和胆酸盐的能力、抑制胰脂肪酶的作用进行分析,评价其体外降脂能力。结果表明,橘皮不溶性膳食纤维的最佳提取条件为提取时间50 min、碱液浓度10%、液料比(mL/g)23:1、提取温度57℃,在此条件下不溶性膳食纤维得率为33.5%。此膳食纤维对大豆油吸附量为3.87 g/g,与胆固醇胶束最大结合能力为4.04 mg/g,对甘氨胆酸钠和牛磺胆酸钠的最大结合率分别为35%和42%,对胰脂肪酶有明显的抑制作用,IC50值(半抑制量)398 mg。橘皮不溶性膳食纤维具有一定的体外降脂能力,有进一步开发降脂功能食品的潜力。The extraction process of insoluble dietary fiber from orange peel residue by alkali method was optimized by response surface test.The ability of the dietary fiber to bind cholesterol micelles and cholate and its inhibitory effect on pancreatic lipase were analyzed to evaluate its lipid-lowering ability in vitro.The results showed that the optimal extraction conditions of insoluble dietary fiber were extraction time 50min,alkali concentration 10%,liquid-solid ratio(mL/g)23:1 and extraction temperature 57℃.Under these conditions,the yield of insoluble dietary fiber was 33.5%.The adsorption capacity of insoluble dietary fiber of orange peel residue to soybean oil was 3.87 g/g.The maximum binding capacity to cholesterol micelles was 4.04 mg/g.The maximum binding rate of insoluble dietary fiber to sodium glycocholate and sodium taurocholate was 35%and 42%,respectively.It had obvious inhibitory effect on pancreatic lipase with IC50value(half inhibitory amount)of 398 mg.These results indicate that insoluble dietary fiber from orange peel has lipid-lowering ability in vitro,there is potential for further development of lipid-lowering functional foods.
