摘 要:【目的】筛选具有抑制黄嘌呤氧化酶(XOD)活性的乳酸菌,为降低家禽尿酸水平提供参考。【方法】测定26株乳酸菌细胞内容物(CFE)对XOD的体外抑制能力,从中筛选出抑制率较高的菌株进行细胞代谢物(CFS)对XOD抑制率的测定,并对其人工胃肠液耐受性、抗菌药物敏感性、安全性等特性进行评价及分子生物学鉴定。【结果】4株乳酸菌A2、A6、SC、SN的CFE对XOD的抑制率分别为23.41%、27.45%、24.47%、27.23%,CFS对XOD的抑制率分别为14.26%、17.02%、22.34%、19.57%。4株乳酸菌经人工胃肠液处理后的活菌数保持在108 CFU·mL^(-1)以上;对头孢菌素类(头孢哌酮、头孢呋辛)、大环内酯类(红霉素、罗红霉素)、青霉素类(阿莫西林、氨苄西林、青霉素)、恶唑烷酮类(利奈唑胺)抗菌药物敏感;基于16S rDNA序列分析,A2菌株鉴定为副干酪乳杆菌,A6菌株鉴定为植物乳杆菌,SC菌株鉴定为短乳杆菌,SN菌株鉴定为鼠李糖乳杆菌;小鼠分别连续腹腔注射A2、A6、SC、SN菌液两周后,未见死亡及器官病变。【结论】成功筛选出能够抑制XOD活性的4株乳酸菌,其具有良好的益生性和安全性。【Objective】Lactic acid bacteria with inhibitory effect on xanthine oxidase(XOD)was screened to provide reference for decrease uric acid level in poultry.【Method】Based on the determined XOD inhibition rate of cell-free extracts(CFE)from 26 lactic acid bacteria strains,the strains with higher inhibition rates were further screened for determining the XOD inhibition rate of cell-free excretory supernatants(CFS),meanwhile,their resistances to artificial gastrointestinal fluid,antimicrobial sensitivity,safety and other characteristics were evaluated and identified by molecular biological methods.【Result】The inhibition rates of CFE towards XOD from strains A2,A6,SC and SN were 23.41%,27.45%,24.47%and 27.23%,respectively,with corresponding CFS having XOD inhibition rates of 14.26%,17.02%,22.34%and 19.57%,respectively.The viable counts of the 4 strains could be maintained above 108 CFU·mL^(-1) after digestion with artificial gastrointestinal fluid.They were sensitive to cephalosporins(cefoperazone and cefuroxime),macrolides(erythromycin and roxithromycin),penicillins(amoxicillin,ampicillin and penicillin),and oxazolidinones(linezolid).Based on 16S rDNA sequence analysis,A2 was identified as Lactobacillus paracasei,A6 was identified as L.plantarum,SC was identified as L.brevis,and SN was identified as L.rhamnosus.After 2 weeks of continuous intraperitoneal injection of the above bacterial solutions,no death or organ lesions were observed in mice.【Conclusion】Four lactic acid bacteria,showing XOD inhibitory effect,were successfully screened,and were accompanied by good probiotic properties and safety.
