摘  要:目的探讨肉豆蔻挥发油的化学成分、壮阳活性、安全性和可能的作用机制。方法①采用气相色谱-质谱联用技术(GC-MS)对肉豆蔻挥发油进行化学成分分析,通过分子对接技术验证主要化学成分与磷酸二酯酶-5(PDE-5)靶点的相互作用。②取Wistar雄性大鼠60只,随机分为空白组、模型组、金匮肾气丸组和肉豆蔻挥发油高、中、低剂量组,每组10只。除空白组外,其他组大鼠均皮下注射氢化可的松建立肾阳虚模型,然后肉豆蔻挥发油高、中、低剂量组分别给予600 mg/kg、300 mg/kg、100 mg/kg肉豆蔻挥发油灌胃,金匮肾气丸组给予金匮肾气丸1.4 g/kg灌胃,空白组和模型组给予等量蒸馏水灌胃,均1次/d,连续灌胃10 d后进行交配能力试验、血清睾酮水平和精液质量测定。③取Wistar雄性大鼠30只,随机分为3组,每组10只,分别给予1000 mg/kg、3000 mg/kg、6000 mg/kg的肉豆蔻挥发油灌胃,观察大鼠的急性毒性和不良反应发生情况。结果肉豆蔻挥发油中共鉴定出75个化合物,其中丁香酚、肉豆蔻醚和4-萜品醇对PDE-5具有显著抑制作用。与空白组比较,模型组大鼠的捕捉潜伏期、交配潜伏期均明显延长(P均<0.05),交配次数明显减少(P<0.05),血清睾酮水平和精子密度、精子存活率、精子活力均明显降低(P均<0.05);与模型组比较,肉豆蔻挥发油各组和金匮肾气丸组大鼠的捕捉潜伏期、交配潜伏期均明显缩短(P均<0.05),肉豆蔻挥发油中、高剂量组和金匮肾气丸组大鼠的交配次数及肉豆蔻挥发油高剂量组、金匮肾气丸组大鼠的捕捉次数均明显增加(P均<0.05),肉豆蔻挥发油中、高剂量组和金匮肾气丸组大鼠血清睾酮水平和精子密度、精子存活率、精子活力均明显升高(P均<0.05)。结论肉豆蔻挥发油能明显提高肾阳虚大鼠的性欲和性能力,增加血清睾酮水平,改善精液质量,且安全,作用机制可能与挥发油中丁香酚、肉豆蔻�Objective It is to explore the chemical composition,aphrodisiac activity,safety,and possible mechanism of action of the volatile oil from nutmeg.Methods①The chemical composition of the volatile oil from nutmeg was analyzed by gas chromatography-mass spectrometry(GC-MS),and the interaction between the main chemical components and the target of phosphodiesterase-5(PDE-5)was verified by molecular docking technology.②Sixty male Wistar rats were randomly divided into blank group,model group,Jin-gui-shen-qi pill group,and high,medium,and low dose groups of nutmeg volatile oil,with 10 rats in each group.The rats of all groups except for the blank group were subcutaneously injected with hydrocortisone to establish models of kidney Yang deficiency.The high,medium,and low dose groups of nutmeg volatile oil were given nutmeg volatile oil 600 mg/kg,300 mg/kg,and 100 mg/kg by gavage,respectively,the Jin-gui-shen-qi pill group was given Jin-gui-shen-qi pill 1.4 g/kg by gavage,the blank group and model group was given were given the same volume of distilled water by gavage,all once daily.After continuous gavage for 10 days,the mating ability of the rats was test,serum level of testosterone and semen quality measurements were detected.③30 male Wistar rats were randomly divided into3 groups,with 10 rats in each group.They were given nutmeg volatile oil by gavage 1000 mg/kg,3000 mg/kg and 6000 mg/kg,respectively.The acute toxicity and adverse reactions of the rats were observed.Results A total of 75 compounds were identified in the volatile oil of nutmeg,among which eugenol,nutmeg ether,and 4-terpineol had significant inhibitory effects on PDE-5.Compared with the blank group,the capture latency and mating latency of the rats in the model group were significantly prolonged(all P<0.05),their mating frequency was significantly reduced(P<0.05),and the serum level of testosterone,sperm density,sperm survival rate,and sperm motility were significantly reduced(all P<0.05).Compared with the model group,the capture latency and mating
