摘 要:以桑叶为原料,采用碱性蛋白酶酶解制备桑叶降胆固醇肽。以多肽质量浓度及胆固醇抑制率为指标,在单因素试验基础上,采用Plackett-Burman与Box-Behnken法优化桑叶降胆固醇肽的酶解工艺。结果表明:最佳酶解工艺为桑叶蛋白质量浓度9.00g/L、加酶量8.0%(以桑叶蛋白质量计)、酶解时间97min、酶解pH7.0、酶解温度48℃。在此条件下,桑叶多肽质量浓度为0.262 mg/mL,胆固醇抑制率为33.27%。Cholesterol-lowering peptides extracted from mulberry leaf were prepared by enzymolysis with alkaline protease.Using the peptide mass concentration and cholesterol inhibition rate as indicators,and the enzymatic hydrolysis process was optimized by Plackett-Burman and Box-Behnken methods based on single factor experiments.The results showed that the optimal enzymatic hydrolysis process conditions were mulberry leaf protein mass concentration 9.00 g/L,enzyme dosage 8.0%(based on the mass of mulberry leaf protein),enzymatic hydrolysis time 97 min,enzymatic hydrolysis pH 7.0,and enzymatic hydrolysis temperature 48 C.Under these conditions,the mass concentration of the mulberry leaf pep-tide was 0.262 mg/mL,and the inhibition rate of cholesterol was 33.27%.
