摘 要:为探索利用豆渣作为工业发酵原料的方法,从霉豆渣中分离3株具有水解蛋白质特性的菌株。通过革兰氏染色、16S rRNA测序、数据库对比构建进化树,分析菌株的亲缘关系;对菌株发酵豆渣后的可溶性蛋白和可溶性糖含量进行分析;以豆渣发酵产物作为原料培养工业菌,并以发酵豆渣作为主要原料,进行产氨基酸发酵。结果表明,3种菌株分别与Bacillus toyonensis、Stenotrophomonas sepilia和Chryseobacterium hispalense亲缘关系最近;菌株发酵中可溶性蛋白的含量最高达4.32 mg/mL,超过未发酵豆渣可溶性蛋白含量的3倍;可溶性糖最大值为5.64 mg/mL,是未发酵豆渣可溶性糖含量的2倍。以豆渣发酵产物作为原料培养毕赤酵母的OD值均未超过1.5,大肠杆菌和谷氨酸棒状杆菌的OD值最高分别为5.0和5.7。精氨酸产量达到15.8 g/L,鸟氨酸产量最高达27.4 g/L。研究结果为豆渣作为工业发酵原料打下理论基础,为豆渣的高值化利用提供了新思路。In order to explore the method of using bean dregs as industrial fermentation raw material.Three strains with hydrolyzed protein were isolated from mold bean residue.The phylogenetic tree was constructed by Gram staining,16S rRNA sequencing and database comparison,and the genetic relationship of strains was analyzed.The contents of soluble protein and soluble sugar in fermented soybean residue were analyzed.The industrial bacteria were cultured with soybean dregs fermentation products,and fermented soybean dregs were used as the main raw materials for amino acid fermentation.The results showed that three strains were closely related to Bacillus toyonensis,Stenotrophomonas sepilia and Chryseobacterium hispalense,respectively.The content of soluble protein was up to 4.32 mg/mL,which was 3 times higher than that of unfermented soybean residue.The maximum value of soluble sugar was 5.64 mg/mL,which was twice the soluble sugar content of unfermented soybean residue.Escherichia coli,Pichia pastoris and Corynebacterium glutamate were cultured with fermented products of soybean residue.OD of Pichia pastoris did not exceed 1.5,while OD values of Escherichia coli and Corynebacterium glutamate were 5.0 and 5.7,respectively.The yield of arginine reached 15.8g/L,and the highest yield of ornithine reached 27.4 g/L.This study offers a theoretical foundation for using soybean residue as medium for industrial fermentation,and provides a new idea for high-value utilization of soybean residue.
