摘 要:为探究低聚果糖(fructo-oligosaccharides,FOS)、低聚半乳糖(galacto-oligosaccharide,GOS)以及FOS与GOS组合物(FOS+GOS)对小鼠脾淋巴细胞增殖活性、辅助型T细胞(helper T cell,Th细胞)分化的影响,以及对Th1/Th2平衡的调节作用,采用FOS、GOS和FOS+GOS分别孵育细胞,通过MTT比色法检测小鼠脾淋巴细胞增殖水平,通过酶联免疫吸附和PCR技术测定Th1、Th2型细胞因子及转录因子T-bet、GAT A-3的表达情况,明确Th细胞分化方向。FOS、GOS和FOS+GOS对小鼠脾淋巴细胞的增殖活性的影响与其添加浓度相关,125μg/mL质量浓度的FOS、GOS和FOS+GOS均能提高脾淋巴细胞增殖活性,其中FOS+GOS对脾淋巴细胞增殖的促进作用最强。在无刀豆蛋白A(concanavalin A,ConA)刺激时,FOS+GOS对脾淋巴细胞增殖活性的促进作用最强;在有ConA刺激下,几种处理对脾淋巴细胞增殖活性的作用差异不显著。GOS和FOS+GOS处理均能够提高Th1型细胞因子IF N-γ的分泌量以及IF N-γ和I L-2 mRNA的表达水平,并能抑制Th2型细胞因子I L-4的分泌量以及I L-5 mRNA的表达水平,增加T-bet/GAT A-3的比率。FOS+GOS与GOS单独处理均具有免疫调节活性,并能调节Th1/Th2平衡,具有潜在的抗过敏功效。The aim of the present study was to investigate the effects of fructo-oligosaccharides(FOS),galacto-oligosaccharide(GOS),and the mixture of FOS and GOS(FOS+GOS)on the proliferation of splenic lymphocytes,as well as the regulation of Th cell differentiation and Th1/Th2 balance.Spleen lymphocytes of mice were incubated with FOS.GOS and FOS+GOS mixture,respectively,with or without Concanavalin A(ConA)stimulation.Spleen lymphocyte proliferation level wasdetected by MTT colorimetric assay.To explore the differentiation direction of Th cell,the expression ofThl and Th2 cytokines and the transcription factors T-bet and GAT A-3 were determined by enzyme linked immunosorbent assay(ELISA)and PCR,respectively.The results showed that the proliferative activities of FOS,GOS and FOS+GOS mixture on mouse splenic lymphocytes were concentration-dependent,and all of them exhibited promoting splenic cell proliferation activity at the concentration of 125μg/mL,with FOS+GOS mixture having the strongest promotion effect on splenic cell proliferation.There was no significant difference among them on the splenic cell proliferation activity under ConA stimulation.FOS+GOS mixture had the strongest effect on the splenic cell proliferation activity without ConA stimulation.The supplementation of GOS and FOS+GOS mixture increased the secretion of Th1-type cytokine IF N-γand the mRNA expression levels of IF N-γand I L-2,and inhibited the expression and secretion levels of Th2-type cytokine-related mRNA including I L-4 and I L-5.In conclusion,both FOS+GOS mixture and GOS possess immunoregulatory activity and regulate Th1/Th2 balance,which deserve them as potential anti-allergic ingredients.
