摘  要:目的 建立高效液相色谱法测定大黄鱼中的柠檬黄、日落黄、苋菜红、胭脂红和亮蓝5种合成着色剂,为水产及其他食品检测提供借鉴。方法 大黄鱼中的合成着色剂用3%氨水-甲醇提取,正己烷脱脂,氮吹浓缩,冷冻离心,CNW Poly-Sery过聚酰胺固相萃取柱净化浓缩,40%甲醇和20%甲酸的水溶液除去天然色素后,经高效液相色谱二极管阵列检测器测定。通过分析合成着色剂的吸收光谱图,选用柠檬黄428 nm、日落黄483 nm、苋菜红521 nm、胭脂红509 nm、亮蓝625 nm特征吸收波长进行定性定量分析。结果 5种合成着色剂在1.0~50.0 mg/L时线性良好,相关系数(r)≥0.999 5,检出限0.008~0.023 mg/kg,定性结果准确,回收率82.5%~98.4%,相对标准偏差(RSD)1.7%~5.3%。结论 该法净化效果好,回收率稳定,一次样品前处理可同时测定5种人工合成着色剂,可提高工作效率,适合基层实验室开展食品中人工合成色素检测工作。Objective To develop a high performance liquid chromatography(HPLC)method for the determination of five synthetic colorants,including Tartrazine,Sunset Yellow,Amaranth,Ponceau 4R and brilliant blue in large yellow croaker,therefore to provide reference for the detection of aquatic products and other foods.Methods Synthetic colorants in large yellow croaker was extracted with 3%ammonia water-methanol and degreased with n-hexane.After nitrogen blowing concentration and refrigerated centrifugation,the extract was purified with a CNW Poly-Sery solid phase extraction cartridge.The synthetic colorants were detected by HPLC/DAD detector after removal of natural pigments by 40%methanol and 20%formic acid solution.By analyzing the absorption spectra of synthetic colorant,the characteristic absorption wavelengths of Tartrazine 428 nm,Sunset Yellow 483 nm,Amaranth 521 nm,Ponceau 4R 509 nm and brilliant blue 625 nm were selected for the qualitative and quantitative analysis.Results The five synthetic colorants showed good linearity at concentration of 1.0-50.0 mg/L with correlation coefficients(r)≥0.9995.The limits of detection(LOD)were 0.008-0.023 mg/kg.The qualitative results were accurate with the recoveries of 82.5%-98.4%and the relative standard deviations(RSD)of 1.7%-5.3%.Conclusion The method had good purification effect,stable recovery rate,and one sample pretreatment can simultaneously determine five kinds of synthetic colorants.It can improve the working efficiency and is suitable for grass-roots laboratories to carry out the detection of synthetic colorants in food.
