摘 要:将禾谷镰刀菌接种小麦,研究不同温度(10、20、30℃)、水活度(0.95、0.98 aw)和培养时间(7、14、21、28、35 d)条件下原型和修饰型脱氧雪腐镰刀菌烯醇(Deoxynivalenol,DON)的产量以及TRI3、TRI5、TRI6和TRI8等关键TRI基因的表达情况,进而分析两者的关系以及受环境因素单一因素和交互作用的影响。结果表明:原型和修饰型DON产生的最适温度和水活度均为20℃和0.98 aw,大部分基因也在该条件下表达量最高。TRI5基因的表达量与DON和15-乙酰基脱氧雪腐镰刀菌烯醇(15-acetyl-deoxynivalenol,15-ADON)的产量显著相关,TRI6和TRI8基因的表达量与DON、3-乙酰基脱氧雪腐镰刀菌烯醇(3-acetyl-deoxynivalenol,3-ADON)和15-ADON的产量显著相关,而TRI3的表达量与4种毒素的产量均无显著相关性,脱氧雪腐镰刀菌烯醇-3-葡萄糖苷(Deoxynivalenol-3-glucoside,D3G)的产量与TRI基因的表达量均无显著相关性。温度、水活度和培养时间单一因素及其交互作用均可显著影响DON的产生和TRI基因的表达,而修饰型DON受大多数环境因素的影响比较显著。本研究结果可为农产品中原型及修饰型DON真菌毒素的监管、防控等提供理论和科学依据。Wheat was inoculated with Fusarium graminearum to study the production of prototypic and modified deoxynivalenol(DON)and the expression of key TRI genes,such as TRI3,TRI5,TRI6,TRI8,etc.,at different temperature(10,20,30℃),water activities(0.95,0.98 aw),and incubation time(7,14,21,28,35 d).Then the relationship between the two,as well as the effects of single and interactive environmental factors were analyzed.The results showed that the optimal temperature and water activity for the production of prototypic and modified DON were both 20℃and 0.98 aw,and most genes were expressed the highest under these conditions.The expression of TRI5 gene was significantly correlated with the production of DON and 15-acetyldeoxynivalenol(15-ADON),and the expression of TRI6 and TRI8 genes was significantly correlated with the production of DON,3-acetyl-deoxynivalenol(3-ADON)and 15-ADON,whereas TRI3 expression was not significantly correlated with the production of the 4 toxins,and deoxynivalenol-3-glucoside(D3G)production was not significantly correlated with the expression of TRI genes.The single factors of temperature,water activity,and cultivation time,as well as their interactions,could significantly affect the production of DON and the expression of TRI genes,while modified DON was more significantly affected by most environmental factors.This study could provide theoretical and scientific basis for the regulation,prevention and control of prototypic and modified DON mycotoxins in agricultural products.
