摘 要:通过固态发酵及液态发酵方式培养亮菌,制备亮菌多糖,比较二者质量差异。结果表明,液态发酵亮菌多糖(LSF-DTP)中多糖含量为60.91%,多糖重均分子量为2.094×10^(4)Da,分子量分布均一度较高。固态发酵亮菌多糖(SSF-DTP)中多糖含量为74.53%,多糖重均分子量为3.053×10^(4) Da,分子量分布广。LSF-DTP、SSF-DTP的单糖均主要由葡萄糖和半乳糖构成;SSF-DTP中甘露糖、木糖及岩藻糖占比高于LSF-DTP,而LSF-DTP中半乳糖醛酸、葡萄糖醛酸、鼠李糖占比高于SSF-DTP。体外抗氧化活性检测结果表明,两者均有明显的抗氧化活性。LSF-DTP的总还原力、DPPH自由基清除率高于SSF-DTP;而在·OH自由基清除率检测中,低浓度时二者活性相当,在质量浓度大于2 mg/mL时,SSF-DTP对·OH自由基清除率更高。此外,LSF-DTP的抗氧化活性及促脾淋巴细胞增殖活性高于SSF-DTP,且分子量较低,均一度更高,液态发酵周期短,更易于质量控制,适用于活性制剂或功能产品的开发。This study compared the quality of polysaccharides derived from Desarmillaria tabescens produced by solid-state fermentation(SSF)and liquid-state fermentation(LSF).The results showed that the polysaccharide content in the liquid-state fermentation of Desarmillaria tabescens-derived polysaccharide(LSF-DTP)was 60.91%,with an average molecular weight of 2.094×10^(4) Da and highly homogeneous molecular weight distribution.In contrast,the solid-state fermentation of Desarmillaria tabescens-derived polysaccharide(SSF-DTP)had a higher polysaccharide content of 74.53%,an average molecular weight of 3.053×10^(4) Da,and a less homogeneous molecular weight distribution.Both LSF-DTP and SSF-DTP were primarily composed of glucose and galactose;however,SSF-DTP contained significantly higher proportions of mannose,xylose,and fucose compared to LSF-DTP,which had higher proportions of galacturonic acid,glucuronic acid,and rhamnose.In vitro experiments revealed that both polysaccharides had notable antioxidant activity.LSF-DTP exhibited significantly higher total reducing power and DPPH radical scavenging activity compared to SSF-DTP.The study results also indicated that both polysaccharides showed similar hydroxyl radical scavenging activity at lower concentrations.However,SSF-DTP had a higher scavenging rate for hydroxyl radicals at concentrations greater than 2 mg/mL.Additionally,LSF-DTP demonstrated a significantly stronger proliferative effect on splenic lymphocytes than SSF-DTP.In conclusion,LSF-DTP has superior antioxidant activity and splenic lymphocyte proliferation stimulation compared to SSF-DTP.With its lower molecular weight,higher uniformity,and shorter fermentation period,LSF-DTP is more suitable for quality control and the development of active preparations or functional products.
