摘 要:为提高斜链虫草(Cordyceps cateniobliqua(Z.Q.Liang)Kepler,B.Shrestha&Spatafora)MB11菌丝体多糖的产量,对斜链虫草的液体深层发酵条件进行优化。采用单因素试验优化了斜链虫草菌丝体多糖发酵的培养基成分,在此基础上,运用Plackett-Burman试验、最陡爬坡试验、Box-Behnken响应面法进一步优化了培养基、接种量、装液量、温度和初始pH值等发酵生产条件。优化后的培养条件如下:蔗糖30 g/L、黄豆匀浆滤液6.84 g/L、MgSO_(4)·7H_(2)O 0.93 g/L、KCl 1 g/L、维生素C 1 g/L、接种量4%、装液量50%、发酵温度18.0℃、摇床转数160 r/min、初始pH6.25。在最优条件下,斜链虫草菌丝体多糖的产量为(703.31±19.93)mg/mL,比优化前提高了3.76倍。In order to enhance the production of mycelial polysaccharides of Cordyceps cateniobliqua(Z.Q.Liang)Kepler,B.Shrestha&Spatafora MB11,its production conditions of Cordyceps cateniobliqua was optimized under submerged liquid fermentation.The medium compositions including carbon and nitrogen sources,inorganic salts,and growth factors were screened by single-factor experiment method.Based on the result of single factor experiment,Plackett-Burman test,steepest ascent test,and Box-Behnken response surface method were successively used to optimize the production condition of mycelial polysaccharides.such as medium,liquid amount,inoculum size,fermentation temperature,and initial pH value.The optimal fermentation condition was obtained as follows:sucrose 30 g/L,soybean homogenate filtrate 6.84 g/L,MgSO_(4)·7H_(2)O 0.93 g/L,KCl 1 g/L,vitamin C 1 g/L,inoculum concentrations 4%,liquid volume 50%,fermented temperature 18.0℃,rotational speed 160 r/min,initial pH 6.25.Under the optimal fermentation condition,the yield of mycelial polysaccharides of Cordyceps cateniobliqua was up to(703.31±19.93)mg/mL,which was 3.76-fold higher than prior to optimization.
