摘 要:以菊花脑为原料,乙醇为提取溶剂,采用超声波辅助法提取菊花脑多酚。通过单因素试验考察乙醇体积分数、料液比、超声时间和超声温度4个因素对菊花脑多酚提取率的影响,通过正交试验得出提取菊花脑多酚的最佳工艺,采用总抗氧化性能、ABTS自由基清除能力、DPPH自由基清除能力和羟自由基清除能力评价菊花脑多酚的抗氧化活性。结果表明,菊花脑多酚的最优提取条件为乙醇体积分数70%、料液比1∶10(g·mL^(-1))、超声时间90 min、超声温度70℃,此时提取率为0.253 mg·g^(-1);菊花脑多酚在浓度为0.015 mg·mL^(-1)时抗氧化性能最强,达132.19 U·mL^(-1),对3种自由基具有良好的清除能力。Polyphenols were extracted by ultrasonic assisted method using Chrysanthemum nankingense as the raw material and ethanol as the extraction solvent.A single factor experiment was conducted to examine the effects of four factors:ethanol volume fraction,solid-liquid ratio,ultrasonic time and ultrasonic temperature on the extraction rate of polyphenol from Chrysanthemum nankingense.The optimal process for extracting Chrysanthemum nankingense polyphenol was obtained through orthogonal test,and its antioxidant activity was evaluated using total antioxidant performance,ABTS free radical scavenging ability,DPPH free radical scavenging ability,and hydroxyl free radical scavenging ability.The results showed that the optimal extraction conditions for Chrysanthemum nankingense polyphenols were:ethanol volume fraction 70%,solid-liquid ratio 1∶10(g·mL^(-1)),ultrasonic time of 90 min,and ultrasonic temperature of 70℃.And under these conditions the extraction rate of Chrysanthemum nankingense polyphenols was 0.253 mg·g^(-1).In addition,Chrysanthemum nankingense polyphenols have the strongest antioxidant activity at a concentration of 0.015 mg·mL^(-1),reaching 132.19 U·mL^(-1),and have good scavenging ability against three types of free radicals.
