摘 要:围绕黄浆水大豆低聚糖含量的测定,将2种快速检测物理方法(手持糖度计和折光仪测定法)与国标法(HPLC法)进行比对,建立具有相关性的线性回归方程,建立低成本和操作简易的大豆低聚糖快检方法。将经过前处理(去蛋白和还原糖)的样品分别使用高效液相色谱法检测出样品中大豆低聚糖的质量浓度,使用折射仪检测其折射率,以及使用糖度计检测浓度百分比。将折射率和浓度百分比作为可变量,质量浓度作为目标量建立线性回归方程。结果表明,使用手持糖度计得出的质量浓度与Brix(g/100 mL)的回归数学模型方程为:黄浆水中低聚糖质量浓度Y=4.348+1.159X+0.2913X^(2),p<0.05,拟合优度可决系数R^(2)接近100%,表示Brix(%)和具体浓度之间的关联在统计意义上显著且该模型有效。使用折光仪得出的质量浓度与折射率的回归方程为:黄浆水中低聚糖质量浓度Y'=1025610-1540504X'+578475X'^(2),p<0.05,R^(2)=100%,表示折射率和质量浓度之间的关联在统计意义上显著且该模型有效。通过验证试验,模型估测值与实际数值之间没有显著差异(p>0.05),说明2种模型可预测黄浆水含低聚糖的质量浓度。二次多项式回归拟合模型能更好地计算黄浆水中大豆低聚糖的实际质量浓度,从而做到以非色谱法的快检方法检测黄浆水中的大豆低聚糖。To establish a linear regression equation with correlation by comparing the detection data of high performance liquid chromatography(HPLC)and rapid detection method,so as to achieve the specific concentration of soybean oligosaccharides in yellow pulp water by rapid detection method.The pretreatment samples were analyzed by high performance liquid ch-romatography(HPLC),refractive index by refractometer,and percentage concentration by saccharimeter.A linear regression equation was established with refractive index and concentration percentage as the variable and specific concentration as the target quantity.The results showed that:The equation of the mathematical model for the regression of specific concentration against Brix(g/100 mL)using the handheld saccharimeter was:Y=4.348+1.159X+0.2913X^(2).The p-value for the linear term Brix and the p-value for the quadratic term Brix2 were less than the significance level of 0.05,indicating that the association between Brix(%).The R^(2) value was 100%,indicating that the model fits the data well.The regression equation of specific concentration and refractive index using refractometer was:Y'=1025610-1540504X'+578475X'^(2).p-values for the linear term refractive index and p-values for the quadratic term refractive index2 were less than the significance level of 0.05,indicating that the association between refractive index and specific concentration was statistically significant and the model was valid.The R^(2) value of 100%indicates that the model fits the data well.Moreover,there was no significant diffe-rence between the model estimates and the actual values(p>0.05),indicating that both model models could predict the concentration of oligosaccharides contained in yellow serofluid.The quadratic polynomial regression fitting model could better calculate the actual concentrations of soybean oligosaccharides in yellow pulp water,thus enabling the detection of soybean oligosaccharides in yellow pulp water by a non-chromatographic method of rapid detection.
