摘  要:通过溶剂提取和大孔树脂纯化得到了云南小粒咖啡果皮多酚(PYCAP),并测定了其生物活性。通过Lineweaver-Burk方程和Dixon方程探究了PYCAP对α-葡萄糖苷酶、α-淀粉酶的抑制机理,分析了其对胰脂肪酶和黄嘌呤氧化酶的抑制作用。采用超高效液相色谱-四极杆飞行时间质谱仪对PYCAP的化学成分进行了鉴定。结果表明,PYCAP具有较强的降血糖、降血脂和降尿酸能力;PYCAP对α-葡萄糖苷酶和α-淀粉酶抑制作用显著,其半数抑制质量浓度(IC_(50))分别为(0.073±0.002)和(0.056±0.005)g/L,且抑制类型均为竞争性-非竞争性混合抑制;PYCAP对胰脂肪酶、黄嘌呤氧化酶均有一定的抑制作用,其IC_(50)分别为(0.034±0.003)和(0.251±0.011)g/L。PYCAP中共鉴定出27种多酚类化合物,主要为黄酮类10种、吡喃类7种和酚酸类10种。Polyphenols from Yunnan Coffee arabica pericarp(PYCAP)was obtained by solvent extraction and macroporous resin purification,and evaluated for their bioactivity.The inhibitory mechanisms of PYCAP onα-glucosidase,α-amylase were analyzed by Lineweaver-Burk equation and Dixon equation,respectively,while the inhibitory effect of PYCAP on pancreatic lipase and xanthine oxidase were evaluated.The chemical composition was further identified by the ultra performance liquid chromatography-quadrupole time of flight mass spectrometry.The results showed that PYCAP exhibited strong antihyperglycemic,hypolipidemic and uric-acid-lowering performance,and significantly inhibitedα-glucosidase andα-amylase,with the corresponding half inhibition mass concentration(IC_(50))of(0.073±0.002)and(0.056±0.005)g/L,and the inhibition types of both competitive-noncompetitive mixed inhibition.In addition,PYCAP inhibited pancreatic lipase and xanthine oxidase to some extent,with the corresponding IC_(50) of(0.034±0.003)and(0.251±0.011)g/L,respectively.A total of 27 chemical components were identified in PYCAP,including 10 flavonoids,7 pyrans,and 10 phenolic acids.
