摘 要:通过单因素和响应面法优化了酶辅助离子液体对大球盖菇粗多糖(SRP)的提取工艺,纯化后得到相对分子质量均一的大球盖菇多糖(SRP-Ⅰ)。通过UV-Vis、FTIR、GC-MS、XRD、SEM、EDS、NMR表征和刚果红实验、TG、部分酸水解、Smith降解测试,探究了SRP-Ⅰ的结构和性质,评价了SRP-Ⅰ的体外抗氧化和降糖能力。结果表明,在提取时间106 min、提取温度51℃、1-十四烷基-3-甲基咪唑溴盐质量浓度4.3 g/L、纤维素酶添加量(以样品质量计,下同)为10.4%的最佳提取工艺条件下,SRP的提取率为32.54%±0.12%;SRP-Ⅰ是1种具有三螺旋构象的α-D-吡喃构型的中性多糖,表面平整光滑,无定形和晶体共存,其在550℃时残炭率为26.34%;SRP-Ⅰ重均相对分子质量为24.321 kDa,主要由葡萄糖(53.31%,摩尔分数,下同)、半乳糖(43.32%)和岩藻糖(3.37%)组成,其中,岩藻糖主要存在于侧链上,葡糖糖和半乳糖较均匀分布在主链和侧链上,主链是以[→3]-Glcp(1→)和[→6]-Galp(1→)结构组成,同时存在[→3,6]-Glcp(1→)支链结构;质量浓度为2.500 g/L SRP-Ⅰ对1,1-二苯基-2-三硝基苯肼自由基、2,2-联氮-双(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基、羟基自由基的清除率分别为82.54%、51.62%、51.57%,对应的半数抑制浓度(IC_(50))分别为0.127、2.438、2.446 g/L,对α-淀粉酶和α-葡萄糖甘酶的抑制率分别为61.2%和71.4%,IC_(50)分别为0.970和0.240 g/L。Stropharia rugosoannulata polysaccharide(SRP-Ⅰ)with uniform relative molecular mass was obtained from enzyme-assisted ionic liquid extraction of Stropharia rugosoannulata after purification,with the process optimized by single factor experiments and response surface analysis,and evaluated via UV-Vis,FTIR,GC-MS,XRD,SEM,EDS,and NMR characterization,Congo Red,TG,partial acid hydrolysis,Smith degradation tests for structure and property analyses.The in vitro antioxidant and hypoglycemic activities of SRP-Ⅰwere further analyzed.The results showed that under the optimal extraction process of extraction time 106 min,extraction temperature 51℃,1-tetradecyl-3-methylimidazolium bromide salt mass concentration 4.3 g/L,cellulase addition(based on the mass of sample,the same below)10.4%,the extraction rate of SRP-Ⅰreached 32.54%±0.12%.SRP-Ⅰwas aα-D-pyran neutral polysaccharide with triple helical conformation,flat and smooth surface,amorphous and crystals coexistence,and a residual carbon rate of 26.34%at 550℃.SRP-Ⅰhaving a weight-average relative molecular mass of 24.321 kDa was mainly composed of glucose(53.31%,mole fraction,the same below),galactose(43.32%)and fucose(3.37%),with fucose mainly present in the side chain,while glucose and galactose more homogeneously distributed in the main chains and side chains.The main chains were mainly in the form of[→3]-Glcp(1→)and[→6]-Galp(1→)structures,while the branched chains were in the form of[→3,6]-Glcp(1→).At SRP-Ⅰmass concentration of 2.500 g/L,the scavenging rates of 1,1-diphenyl-2-trinitrophenylhydrazine radicals,2,2-diazo-di(3-ethylbenzothiazole-6-sulfonic acid)diamine salt radicals and hydroxyl radicals were up to 82.54%,51.62%and 51.57%,the corresponding median inhibitory concentration(IC_(50))were 0.127,2.438 and 2.446 g/L,the inhibition rates ofα-amylase andα-glucosidase were 61.2%and 71.4%,and the IC_(50) were 0.970 and 0.240 g/L,respectively.
