摘 要:本研究以实验室前期通过常压室温等离子体(ARTP)诱变所得一株高产DHA裂殖壶藻突变株I-F-9及原始藻株ATCC20888为研究对象,评价ARTP诱变前后裂殖壶藻生物活性物质及氨基酸的营养价值。利用全自动氨基酸分析仪测定裂殖壶藻中氨基酸的含量后,经氨基酸评分(AAS)、氨基酸比值(RAA)、氨基酸比值系数(RC)、比值系数分(SRC)等营养学方法对藻株进行营养价值评价。结果表明:在生物活性物质方面,突变株I-F-9的维生素E、β-胡萝卜素、多糖、总糖和虾青素含量显著高于原始藻株(P<0.05)。其中突变株I-F-9的维生素E、β-胡萝卜素、多糖、总糖和虾青素含量分别为18.80μg/g、517.85μg/g、76.18 mg/g、234.74 mg/g、1833.47 pg/g,比原始株分别提高了90.28%、59.29%、19.76%、71.89%、5.18%。原始株与裂殖壶藻突变株I-F-9含17种氨基酸,其中突变株I-F-9苏氨酸、缬氨酸、赖氨酸3种必需氨基酸均显著高于原始株(P<0.05)。突变株I-F-9非必需氨基酸、总氨基酸含量均显著高于原始株(P<0.05)。营养价值评价结果表明,突变株I-F-9与原始株的第一限制氨基酸为赖氨酸,其中突变株I-F-9 SRC值为83.91,原始株的SRC值为84.19。ARTP诱变显著提升了裂殖壶藻突变株维生素E、β-胡萝卜素、多糖、总糖和虾青素含量及非必需氨基酸、总氨基酸含量。In this study,a high-yield DHA Schizochytrium mutant strain I-F-9 and the original strain ATCC20888 obtained by atmospheric room temperature plasma(ARTP)mutagenesis in the laboratory were used as the research objects to evaluate the bioactive substances and amino acid nutritional value of Schizochytrium before and after ARTP mutagenesis.After the content of amino acids in Schizochytrium was determined by automatic amino acid analyzer,the nutritional value of the algae was evaluated by amino acid score(AAS),amino acid ratio(RAA),amino acid ratio coefficient(RC),ratio coefficient score(SRC)and other nutritional methods.The results showed that the contents of vitamin E,β-carotene,polysaccharide,total sugar and astaxanthin in mutant I-F-9 were significantly higher than those in the original strain(P<0.05).The contents of vitamin E,β-carotene,polysaccharide,total sugar and astaxanthin in the mutant strain I-F-9 were 18.80μg/g,517.85μg/g,76.18 mg/g,234.74 mg/g and 1833.47 pg/g,respectively,which were 90.28%,59.29%,19.76%,71.89%and 5.18%higher than those of the original strain.The original strain and Schizochytrium sp.mutant strain I-F-9 contained 17 amino acids,among which the three essential amino acids of threonine,valine and lysine in the mutant strain I-F-9 were significantly higher than those in the original strain.(P<0.05).The contents of non-essential amino acids and total amino acids in I-F-9 were significantly higher than those in the original strain.The contents of non-essential amino acids and total amino acids in I-F-9 were significantly higher than those in the original strain(P<0.05).The results of nutritional value evaluation showed that the first limiting amino acid of the mutant strain I-F-9 and the original strain was lysine,in which the SRC value of the mutant strain I-F-9 was 83.91,and the SRC value of the original strain was 84.19.ARTP mutagenesis significantly increased the contents of vitamin E,β-carotene,polysaccharides,total sugar and astaxanthin,as well as the contents of non-essential am
