摘 要:为获得高产细菌素菌株,对高产突变菌株进行全基因组测序分析,以探究菌株高效产细菌素的分子机制。将原始菌株戊糖片球菌C-2-1进行多轮紫外诱变处理,突变菌株利用二代测序平台进行测序,然后进行组装和功能注释。组装得到的全基因组序列与原始菌株进行比较基因组学分析。结果表明,经过8轮紫外诱变获得一株正突变菌株C282,其抑菌效价1371.50 IU·mL^(-1),相较原始菌株(170.81IU·mL^(-1))提高7.03倍。菌株C282的基因组是由一个1976069 bp的染色体组成,GC含量为37.13%,共编码2049个蛋白。全基因组的共线性分析说明菌株C282基因组与原始菌株C-2-1具有显著差异;与原始菌株相比,C282菌株新增了一种编码细菌素pediocin PA-1的次级代谢产物合成基因簇,推测是原始菌株对环境变化的响应;通过GO、KEGG、COG数据库比对C282和原始菌株的基因组,发现其两株菌的差异基因主要富集在碳水化合物代谢和氨基酸代谢等相关通路。该研究揭示了原始菌株与突变菌株的差异基因信息,为原始菌株的定点改良确定了基因靶点,为比较基因组学改良其他菌株提供了范式。To obtain a high bacteriocin-producing strain,whole genome sequencing analysis was conducted on the mutant strain to explore the molecular mechanism of efficient bacteriocin production by the strains.The original strain Pediococcus pentosaceus C-2-1 was subjected to multiple rounds of UV mutagenesis,and the mutant strains were sequenced using a second-generation sequencing platform,followed by genome assembly,gene prediction and functional annotation.The whole genome sequences obtained from the assembly were subjected to com-parative genomic analysis with the original strain.The results showed that a positive mutant strain C282 was obtained after 8 rounds of UV mutagenesis,and its inhibitory potency was 1371.50 IU·mL^(-1),which was 7.03 times higher than that of the original strain(170.81 IU·mL^(-1)).The genome of strain C282 was composed of a 1976069 bp chromosome with 37.13%GC content,encoding a total of 2049 proteins.The genome-wide covariance analysis indicated that the genome of strain C282 was significantly different from that of the original strain C-2-1;compared with the original strain,strain C282 added a new gene cluster for sec-ondary metabolite synthesis encoding bacteriocin pediocin PA-1,which was presumed to be the response of the original strain to environmental changes;the GO,KEGG,and COG data-bases were used to compare the genomes of C282 and the original strain,and it was found that the genes of the two strains were mainly enriched in carbohydrate and amino acid metabolism pathways.The study revealed the genetic information of the original strain and the mutant strain,which identified the gene targets for the targeted improvement of the original strain and provided a paradigm for the improvement of other strains by comparative genomics.
