摘 要:优化连翘三萜酸的超声辅助提取工艺,并对其进行定性分析和抗氧化活性评价。通过单因素实验及Box-Behnken设计,对连翘三萜酸的超声辅助提取工艺进行优化;利用高效液相色谱(HPLC)对三萜酸进行定性分析;采用多种自由基清除法和铁离子还原能力(FRAP)法评估其抗氧化活性。结果表明:连翘三萜酸超声辅助最佳提取工艺为乙醇质量分数80%、料液比25(g/mL)、超声时间62 min、超声温度50℃,在此条件下,连翘三萜酸得率为15.52%,与理论预测值(15.58%)相对误差为0.39%。HPLC定性结果显示,连翘三萜酸中含有熊果酸、白桦脂酸和齐墩果酸。连翘三萜酸提取物具有较强的清除DPPH、ABTS+、·OH自由基能力和还原Fe3+的能力,其抗氧化活性与质量浓度呈现出量效关系,且其抗氧化活性整体优于单一的熊果酸、白桦脂酸或齐墩果酸。优化后的连翘三萜酸超声辅助提取工艺稳定可行,且其提取物具有较强抗氧化活性。The ultrasonic-assisted extraction(UAE) of triterpenoid acids from Forsythia suspensa was optimized using single-factor experiments and Box-Behnken design.Qualitative analysis of triterpenoid acid was conducted using high performance liquid chromatography(HPLC) and antioxidant activity was evaluated using free radical scavenging methods and Ferric Reducing Antioxidant Power(FRAP) assay.The results indicated that the optimal ultrasonic-assisted extraction conditions for triterpenoid acid from Forsythia suspensa was as follows:an ethanol volume fraction of 80%,a solid-to-liquid ratio of 25(g/mL),an ultrasonic time of 62 minutes,and an ultrasonic temperature 50℃.Under the above conditions,the yield of triterpenoid acids was 15.52%,with a relative error of 0.39% compared to the theoretical predicted value(15.58%).The HPLC analysis revealed the presence of ursolic acid,betulinic acid and oleanolic acid in triterpenoid acids from Forsythia suspensa.The extract exhibited potent antioxidant activity,effectively scavenging DPPH,ABTS+,and · OH free radicals,and reducing Fe3+ in a concentration-dependent manner.Its antioxidant activity surpassed that of the individual triterpenoid acids,such as ursolic acid,betulinic acid,or oleanolic acid.This optimized UAE method was found to be stable and feasible,and its extract demonstrated strong antioxidant activity.
