摘 要:针对木薯酒精清液发酵中的产朊假丝酵母培养条件进行了优化。通过调整溶解氧水平和补料时间点,显著提高了酵母的生长效率和菌体蛋白的质量。结果表明:在溶解氧水平约为10%时,酵母生长表现最佳,发酵终点酵母浓度可达7.9亿个·mL^(-1)。同时,选择在第10 h进行补料,能够有效维持底物糖浓度,促进酵母持续生长,最终酵母数量达到11.80亿个·mL^(-1)。此外,优化后的条件还显著提高了发酵液中的蛋白质质量分数,达到49.01%。7 L罐发酵实验进一步验证了这些优化条件的可行性,菌体蛋白的绝干收率达40.4 g·L^(-1)发酵液,显示出其在工业化生产中的潜力。The cultivation conditions for Pichia pastoris in cassava alcohol distillery wastewater fermentation were optimized.By adjusting the dissolved oxygen levels and feeding time points,the growth efficiency of the yeast and the quality of the yeast protein were significantly improved.Experimental results indicated that dissolved oxygen level of approximately 10%provided the best growth conditions,with final yeast concentration reaching 790 million cells per milliliter.Additionally,feeding at the 10-hour mark effectively maintained substrate sugar concentration,promoting continued yeast growth,resulting in a final yeast concentration of 1.18 billion cells per milliliter.The optimized conditions also significantly increased the protein mass fraction in the fermentation broth,reaching 49.01%.A 7 L fermenter experiment further validated the feasibility of these optimized conditions,with the dry weight yield of yeast protein reaching 40.4 g·L^(-1) of fermentation broth,demonstrating its potential for industrial production.
