摘  要:为高效制备ω-3型中长链甘油酯,以α-亚麻酸和椰油为原料进行酶促酯交换。以产物中α-亚麻酸的含量为指标,筛选出3种固定化脂肪酶进行混料试验,确定酶最佳质量比;在此基础上,采用响应面法优化ω-3型中长链甘油酯制备工艺,并对复合酶的稳定性进行分析。结果表明:Lipozyme RM C、Lipozyme TL TM和Novozyme 435酯交换效果较好,最佳质量比为0.30∶0.25∶0.45;最优工艺参数为底物质量比5.5∶1、加酶量5.3%(以底物总质量计)、温度50.0℃。在此条件下得到的α-亚麻酸含量为44.87%±1.41%,制得的复合酶具有良好的稳定性。In order to prepareω-3 type of medium-long chain glycerides efficiently,α-linolenic acid and coconut oil were used as raw materials for enzymatic transesterification.Taking the content of linolenic acid in the product as an indicator,3 immobilized lipases were selected out for mixing experiments.A regression model was constructed to determine the optimal enzyme mass ratio.On the basis,response surface methodology was used to optimize the preparation process ofω-3 medium long chain glycerides,and the stability of the composite enzyme was analyzed.The results showed that Lipozyme RM C,Lipozyme TL TM and Novozyme 435 exhibited better transesterification effect,with the optimal mass ratio being 0.30∶0.25∶0.45.The optimal process parameters were substrate mass ratio 5.5∶1,enzyme dosage 5.3%(based on the total substrate mass),and temperature 50.0℃.The linolenic acid content obtained under the optimal conditions was 44.87%±1.41%,and the obtained compound enzyme had good stability.
