摘 要:采用碘-淀粉显色法建立了α-淀粉酶抑制剂的筛选方法。对淀粉浓度、α-淀粉酶浓度、有机溶剂甲醇的用量和反应终止液盐酸浓度进行了逐一考察,结果表明最佳测定条件淀粉基质液质量浓度为0.0035 g·mL^(-1),淀粉酶基质液质量浓度为0.05 mg·mL^(-1),有机溶剂甲醇的体积分数不超过10%为宜,反应终止液盐酸浓度选用1.0 mol·L^(-1)最佳。以阿卡波糖为阳性对照,比较了对香豆酸、儿茶素和岩白菜素对α-淀粉酶的抑制活性,同浓度时对香豆酸对α-淀粉酶的抑制活性最强,并测得对香豆酸的IC_(50)值为2.79 mg·mL^(-1)。A screening method for α-amylase inhibitors was established based on the iodine-starch coloritmetry.Starch concentration,α-amylase concentration,the concentrations of organic solvent methanol and hydrochloric acid for stopping reaction were investigated one by one.The optimal determination conditions were described as follows:the concentration of starch matrix solution was 0.0035 g·mL^(-1),the concentration of amylase matrix solution was 0.05 mg·mL^(-1),the concentration of organic solvent methanol was not more than 10%,and the concentration of hydrochloric acid for terminated action was 1.0 mol·L^(-1).The inhibitory activity of p-coumaric acid,catechin and bergenin on α-amylase were investigated with the acarbose as the positive control drug,the results showed that the inhibitory activity of p-coumaric acid onα-amylase was the highest one,and the IC_(50) value of p-coumaric acid was 2.79 mg·mL^(-1).
