摘  要:目的:通过斑马鱼模型辅助评估植物原料降糖活性。方法:评价4种植物提取物的体外抑制α-葡萄糖苷酶的活性,并以麦芽糊精为升糖诱导剂建立斑马鱼高糖模型,评价植物提取物的降糖效果。结果:体外酶活实验显示,表没食子儿茶素没食子酸酯(来源于绿茶)、桑叶提取物、绿茶提取物和白芸豆提取物对α-葡萄糖苷酶的半抑制浓度(IC_(50))分别为0.9927、11.54、53.82μg·mL^(-1)和9035μg·mL^(-1)。在斑马鱼高糖模型下,1 mg·mL^(-1)表没食子儿茶素没食子酸酯和1 mg·mL^(-1)桑叶提取物可以降低麦芽糊精诱导的斑马鱼体内的葡萄糖含量,分别降低64.40%和68.56%,具有显著性差异(p<0.01)。结论:本文建立的斑马鱼评价模型可用于筛选具有降糖活性的植物提取物,具有潜在的应用前景。Objective:To evaluate hypoglycemic activity of plant raw materials by zebrafish model.Method:The inhibitory activity of 4 plant extracts onα-glucosidase in vitro was evaluated,and the high glucose model of zebrafish was established with maltodextrin as glucose-raising inducer to evaluate the hypoglycemic effect of plant extracts.Result:The semi-inhibitory concentrations(IC_(50))of epigallocatechin gallate(derived from green tea),mulberry leaf extract,green tea extract and white kidney bean extract onα-glucosidase were 0.9927,11.54,53.82μg·mL^(-1) and 9035μg·mL^(-1),respectively.In zebrafish high-glucose model,1 mg·mL^(-1) epigallocatechin gallate and 1 mg·mL^(-1) mulberry leaf extract could reduce the glucose content of maltodextrin-induced zebrafish by 64.40%and 68.56%,respectively,with significant difference(p<0.01).Conclusion:The zebrafish evaluation model established in this paper can be used for screening plant extracts with hypoglycemic activity,and has potential application prospects.
