八月瓜酵素制备工艺及抗氧化和降血糖活性评价

摘  要:文章以八月瓜为原料,采用单因素试验结合响应面试验优化八月瓜酵素的发酵工艺,得到风味宜人、滋味协调、品质良好的八月瓜酵素,检测了八月瓜酵素中功效成分及风味成分,并评价了其抗氧化活性和降血糖活性。试验结果显示,八月瓜酵素最佳发酵工艺条件为糖添加量11.6%、发酵剂用量0.80%、发酵温度25.3℃、发酵时间17d。抗氧化活性评价表明,当浓度为12.5mg/mL时,八月瓜酵素对1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基清除率为87.7%,此外,其超氧化物歧化酶(Superoxide dismutase,SOD)活力为746.3U/mL。体外降血糖活性试验结果表明,在浓度为2.5~15 mg/mL范围内,八月瓜酵素对α-葡萄糖苷酶和α-淀粉酶抑制率呈剂量依赖性。当浓度为12.5 mg/mL时,八月瓜酵素对α-葡萄糖苷酶的抑制率为78.4%;当浓度为15 mg/mL时,其对α-淀粉酶的抑制率为85.5%。研究结果表明,八月瓜酵素具有较好的抗氧化、降血糖活性,可为八月瓜的进一步开发利用提供科学依据。This study used Akebia trifoliata as raw material and optimized the fermentation process of Akebia trifoliata fermentation liquor using single-factor combined with response surface experiment.A pleasant flavor,harmonious taste,and good quality Akebia trifoliata fermentation liquor was obtained.The efficacy components and flavor components of Akebia trifoliata fermentation liquor were detected,and its antioxidant and hypoglycemic activities were evaluated.The experimental results showed that the optimal fermentation conditions for Akebia trifoliata fermentation liquor were as follows:sugar addition 11.6%,fermentation agent 0.80%,fermentation temperature 25.3℃,and time of 17 d.The evaluation of antioxidant activity showed that when the concentration was 12.5 mg/mL,the 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging rate of Akebia trifoliata fermentation liquor was 87.7%,in addition,the superoxide dismutase(SOD)activity was 746.3 U/mL.The results of in vitro hypoglycemic activity experiment showed that within the concentration range of 2.5–15 mg/mL,the inhibition rates of Akebia trifoliata fermentation liquor onα-glucosidase andα-amylase were dose-dependent.When the concentration was 12.5 mg/mL,the inhibition rate of Akebia trifoliata fermentation liquor onα-glucosidase was 78.4%.When the concentration was 15 mg/mL,the inhibition rate of Akebia trifoliata fermentation liquor onα-amylase was 85.5%.This indicates that Akebia trifoliata fermentation liquor has good antioxidant and hypoglycemic activities.The results of this study can provide scientific basis for the further development and utilization of Akebia trifoliata.
