摘 要:石榴皮是石榴的加工副产物。为促进石榴皮的开发利用,以石榴皮为原料,采用超声辅助酶法提取石榴皮多酚,并对其抗氧化活性进行探究。通过不同溶剂、Plackett-Burman试验、最陡爬坡试验和Box-Behnken响应面法优化石榴皮的多酚提取工艺,并对其抗氧化活性进行评价。结果表明,50%丙酮为最佳提取溶剂,Plackett-Burman试验确定石榴皮多酚提取工艺中影响显著的3个因素:液料比、复合酶比例和酶解p H值。Box-Behnken试验和响应面分析优化提取条件为液固比21:1(mL/g)、复合酶比例(果胶酶:纤维素酶)8:25、酶解pH值为4.6,此时多酚含量为221.81 mg/g。该优化工艺可靠,具有实际可行性。石榴皮多酚的总还原力和浓度在一定范围内成正比,并且具备1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基和2,2'-联氮-双(3-乙基苯并噻唑啉-6-磺酸)(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基清除能力,其IC50分别为5.49μg/mL和9.23μg/mL。体外抗氧化试验结果表明石榴皮多酚具有较强的抗氧化性。Pomegranate peel is a by-product of pomegranate processing.In order to promote the development and utilization of pomegranate peel,pomegranate peel polyphenols were extracted from pomegranate peel by ultrasound-assisted enzymatic extraction,and its antioxidant activity was studied.The extraction process of polyphenols from pomegranate peel was optimized by different solvents,PlackettBurman design,path of steepest ascent method,and Box-Behnken response surface method,and its antioxidant activity was evaluated.The results showed that 50%acetone was the best extraction solvent,and Plackett-Burman test determined three significant factors in the extraction process of polyphenols from pomegranate peel:liquid-material ratio,compound enzyme ratio and enzymatic hydrolysis pH.The optimum extraction conditions of Box-Behnken test and response surface analysis were as follows:liquidsolid ratio 21:1(mL/g),compound enzyme ratio(pectinase:cellulase)8:25,enzymatic hydrolysis pH4.6,and the content of polyphenols was 221.81 mg/g.The optimization process was reliable and feasible.The total reducing power of polyphenols from pomegranate peel was proportional to the concentration in a certain range.And it had the ability of scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2'-azinobis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)free radicals.Its IC50 was 5.49μg/mL and 9.23μg/mL,respectively.The results of antioxidant test in vitro showed that pomegranate peel polyphenols had strong antioxidant activity.
