摘 要:以树莓干果为原料,通过闪式提取树莓干果多酚。采用单因素试验分析了液固比、乙醇体积分数、提取电压、提取次数、提取时间对树莓干果多酚提取率的影响。通过Box-Behnken试验设计优化最佳提取工艺条件,同时考察树莓干果多酚体外抗氧化和降血糖活性。结果表明,最佳提取条件为液固比33:1 mL/g、乙醇体积分数57%、提取电压46 W、提取次数2次、提取时间20 s,得到多酚得率为1.55 mg/g。树莓多酚对1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH)自由基、2,2'-联氮-双(3-乙基苯并噻唑啉-6-磺酸)(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS)阳离子自由基、羟自由基最大清除率和总还原能力分别为93.85%、73.08%、91.04%和0.81,且具有良好的量效关系,其半数抑制浓度(IC_(50))值分别为12.27、52.92、19.04μg/mL和46.08μg/mL。树莓多酚对α-葡萄糖苷酶和α-淀粉酶活性的最大抑制率分别为81.23%和85.25%,其IC_(50)分别为49.36μg/m L和14.36μg/m L。研究结果表明,树莓多酚具有一定的抗氧化和降血糖能力。Polyphenols in dried raspberries were extracted using flash technology.The effects of liquidsolid ratio,ethanol concentration,extraction voltage,extraction times and extraction time on polyphenols extraction rate of dried raspberries were analyzed by single factor experiments.The optimal extraction conditions were optimized by response surface method(RSM),and dried raspberries polyphenols was further analyzed for their antioxidant and hypoglycemic activities in vitro.The results showed that the optimal conditions for the extraction of polyphenols are liquid-solid ratio 33:1 mL/g,ethanol concentration 57%,extraction voltage 46 W,extraction times 2 times and extraction time 20 s.Under these conditions,the extraction yield of polyphenols was 1.55 mg/g.The maximum rate of scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)(ABTS)radical cation,hydroxyl radical and total reducing power were 93.85%,73.08%,91.04%and 0.81,and it has good dose-effect relationship.The half inhibitory concentration(IC_(50))were 12.27,52.92,19.04μg/mL and 46.08μg/mL,respectively.The maximum inhibitory rates of polyphenols in raspberry forα-glucosidase andα-amylase activities were 81.23%and 85.25%,and IC_(50)were 49.36μg/mL and 14.36μg/mL,respectively.These indicated that the polyphenols of raspberry have certain antioxidant and hypoglycemic capacity.
