摘  要:目的:探究不同盐碱地菊芋种质资源菊糖含量的差异性,以及菊芋提取物对Ⅱ型糖尿病(Type 2 diabetes mellitus,T2DM)大鼠糖脂代谢的影响。方法:以高脂饲料联合链脲佐菌素注射诱导建立T2DM大鼠模型,造模成功后随机分为糖尿病模型组、盐酸二甲双胍阳性对照组、菊芋提取物低剂量组(200 mg/kg)、菊芋提取物中剂量组(400 mg/kg)及菊芋提取物高剂量组(800 mg/kg),并设正常对照组。监测大鼠在灌胃期的空腹血糖值、体质量变化,并在干预28 d后,评价各组大鼠口服葡萄糖耐量、胰岛素及糖脂代谢水平。结果:BY1在30个菊芋种质资源中菊糖含量最高,达(196.35±3.25)mg/g,作为试验材料进一步验证其功能。与模型组相比,菊芋试验组大鼠体质量均显著性提高(P<0.05);空腹血糖值均显著降低(P<0.05);血清胰岛素水平显著增加(P<0.05),胰岛素抵抗指数显著下降(P<0.05);口服葡萄糖耐量显著提高(P<0.05);同时,通过下调T2DM大鼠血清中总胆固醇、甘油三酯、低密度脂蛋白胆固醇水平(P<0.05),上调高密度脂蛋白胆固醇水平(P<0.05)以改善血脂异常,尤其以菊芋提取物高剂量组(800 mg/kg)降低血脂水平效果最为明显,与阳性对照组效果相似。结论:菊芋提取物对T2DM大鼠具有显著的降血糖作用,其调控作用可能与维护糖耐量、促进胰岛素分泌和改善血脂代谢有关。Objective:To explore the difference of inulin content in Jerusalem artichoke germplasm resources in saline alkali land,and the effect of Jerusalem artichoke extract on glucose and lipid metabolism in type II diabetes mellitus(T2DM)rats.Methods:T2DM rats were induced by high-fat diet combined with streptozotocin(STZ)injection.After successful modeling,they were randomly divided into diabetes model group,metformin hydrochloride positive control group,Jerusalem artichoke extract low dose group(200 mg/kg),Jerusalem artichoke extract medium dose group(400 mg/kg)and Jerusalem artichoke extract high dose group(800 mg/kg),and set up normal control group.The fasting blood glucose and body weight changes of rats during the gavage period were monitored,and the oral glucose tolerance,insulin,and glucose and lipid metabolism levels of each group of rats were evaluated after 28 days of intervention.Results:BY1 had the highest inulin content among 30 germplasm resources of Jerusalem artichoke,reaching(196.35±3.25)mg/g,which was used as experimental material for further functional verification.Compared with the model group,the body weight of rats in the Jerusalem artichoke experimental group was significantly improved(P<0.05).The fasting blood glucose levels were significantly reduced(P<0.05).The serum insulin level significantly increased(P<0.05),and the insulin resistance index significantly decreased(P<0.05).Oral glucose tolerance significantly increased(P<0.05).At the same time,by downregulating the levels of total cholesterol(TC),triglyceride(TG),and low density lipoprotein cholesterol(LDL-C)in the serum of T2DM rats(P<0.05)and upregulating the level of high density lipoprotein cholesterol(HDL-C)(P<0.05),blood lipid abnormalities were improved,especially in the high-dose group of Jerusalem artichoke extract(800 mg/kg),which showed the most significant effect in reducing blood lipid levels,similar to the positive control group.Conclusion:Jerusalem artichoke extract has a significant hypoglycemic effect on T2DM rats,and its
