摘  要:为了探究番茄红素(LYC)和烟酰胺单核苷酸(NMN)稳定性以及对急性肠损伤的保护作用,在制备LYC脂质体基础上,利用多孔微凝胶共包载LYC脂质体和NMN,然后考察LYC和NMN的抗氧化性及其对细胞炎症和氧化应激的影响,进一步研究LYC和NMN微凝胶对脂多糖(LPS)诱导的C57BL/6小鼠急性肠损伤的保护作用及其潜在机制。结果表明,在体外,番茄红素对于DPPH自由基清除率IC_(50)为111.36μg/mL,对于ABTS自由基清除率IC_(50)为95.66μg/mL,对于羟自由基清除率IC_(50)为73.01μg/mL,微凝胶能够提高番茄红素对自由基的清除能力,并且LYC和NMN通过抑制NO和炎症因子的分泌、恢复细胞SOD酶活力和抑制MDA生成,降低LPS诱发的炎症反应,减轻氧化应激。相关机制研究表明负载LYC和NMN的微凝胶可通过上调ZO-1及Occludin基因表达,显著改善LPS诱导的肠道损伤。To investigate the stability of lycopene(LYC)and nicotinamide mononucleotide(NMN)and their protective effects on acute intestinal injury,based on the preparation of LYC liposomes,this article uses porous microgel to co-encapsulate LYC liposomes and NMN,and then examines the antioxidant properties of LYC and NMN and their effects on cellular inflammation and oxidative stress.Further research is conducted on the protective effects of LYC and NMN microgel on lipopolysaccharide(LPS)-induced acute intestinal injury in C57BL/6 mice and their potential mechanisms.The results show that the ICso oflycopene for DPPH free radical scavenging in vitro is 111.36μg/mL,the ICso for ABTS free radical scavenging is 95.66μg/mL,and the ICso for hydroxyl radical scavenging is 73.01μg/mL.The microgel can improve the scavenging ability of lycopene on free radicals,and LYC and NMN can reduce the inflammatory response induced by LPS and alleviate oxidative stress by inhibiting the secretion of NO and inflammatory factors,restoring the activity of SOD enzyme,and inhibiting the production of MDA.The research on the relevant mechanisms has shown that microgel loaded with LYC and NMN can significantly improve LPS-induced intestinal injury by upregulating the expression of ZO-1 and Occludin genes.
