摘 要:通过体外酶抑制实验和小鼠实验,评估分离纯化后发芽糙米酚类物质的降血糖活性。以发芽糙米为原料,经超声加酶法提取得到发芽糙米多酚粗提物,采用柱层析分离纯化,即得到发芽糙米酚类物质(GBRP)。以α-葡萄糖苷酶和α-淀粉酶活性抑制实验评价体外降血糖活性,并通过高脂饲料和链脲佐菌素联合诱导建立Ⅱ型糖尿病小鼠模型,将实验小鼠分为空白组、模型组、阳性组、GBRP低剂量组(GBRP-L)、GBRP中剂量组(GBRP-M)和GBRP高剂量组(GBRP-H),连续灌胃28 d,测定小鼠的血糖水平,并记录其他相关生理指标。结果显示,发芽糙米酚类物质具有良好的酶抑制率,并显著降低了糖尿病小鼠的空腹血糖,改善了小鼠糖耐量异常。此外,有效地降低了小鼠血清总胆固醇、甘油三酯及低密度脂蛋白胆固醇,增加了高密度脂蛋白胆固醇以及肝脏抗氧化应激和肝细胞保护等功能。The hypoglycemic activity of germinated brown rice phenols was assessed through isolation and purification,using an enzyme inhibition test in vitro and a mouse test.Germinated brown rice was utilized as the raw material to obtain crude polyphenol extract via ultrasonic enzymatic method.The germinated brown rice polyphenol compounds(GBRP)were then obtained through column chromatography.In vitro hypoglycemic activity was assessed byα-glucosidase andα-amylase inhibitory tests,while a type 2 diabetes mouse model was induced using high fat diet combined with streptozotocin.Experimental mice were divided into blank group,model group,positive group,low GBRP dose group(GBRP-L),medium GBRP dose group(GBRP-M),and high GBRP dose group(GBRP-H).Blood glucose levels of the mice were measured continuously for 28 days via gavage,with other related physiological indexes being recorded.Results indicated that germinated brown rice phenols exhibited significant enzyme inhibition rate,leading to a marked reduction in fasting blood glucose levels and improved glucose tolerance in diabetic mice.Furthermore,serum total cholesterol,triglyceride and low-density lipoprotein cholesterol levels were effectively reduced,and the functions of high-density lipoprotein cholesterol,liver antioxidant stress and hepatocyte protection,etc.were increased.
