摘  要:为充分利用桑葚果渣中的花青素,在超声辅助酶提取条件下,以桑葚果渣为原料,探索了料液比、酶用量、超声时间和超声温度4个单因素对桑葚果渣花青素提取量的影响,并采用响应面法优化了超声辅助酶提取花青素工艺,进一步使用超高效液相色谱-质谱联用仪(UPLC-MS)对提取液进行了花青素成分确定。结果表明,花青素最佳提取条件为料液比1∶20、酶用量0.3%、超声时间1.7 h、超声温度55℃,此时桑葚果渣花青素提取量可达7.0712 mg/g。提取液中可检测到10种花青素,主要为矢车菊素-3-O-芸香糖苷(44.56%)、矢车菊素-3-O-芸香糖苷-葡萄糖苷(25.84%)、矢车菊素-3-O-木糖苷(18.41%)和矢车菊素-3,5-O-二葡萄糖苷(7.34%)。该试验结果可为桑葚果渣利用和花青素产品生产提供思路和参考依据。In order to make full use of anthocyanins in mulberry pomace,under the extraction conditions of ultrasonic-assisted enzyme,with mulberry pomace as the raw material,the effects of four single factors such as solid-liquid ratio,enzyme dosage,ultrasonic time and ultrasonic temperature on the extraction amount of anthocyanins from mulberry pomace are investigated.Response surface method is used to optimize the extraction process of anthocyanins with ultrasonic-assisted enzyme,and the anthocyanin components of the extraction solution are determined by UPLC-MS.The results show that the optimal extraction conditions are as follows:the solid-liquid ratio is 1∶20,the enzyme dosage is 0.3%,the ultrasonic time is 1.7 h and the ultrasonic temperature is 55℃.At this time,the extraction amount of anthocyanins from mulberry pomace could reach 7.0712 mg/g.A total of 10 kinds of anthocyanins are detected in the extraction solution,mainly are cyanidin-3-O-rutinoside(44.56%),cyanidin-3-O-rutinoside-glucoside(25.84%),cyanidin-3-O-xyloside(18.41%)and cyanidin-3,5-O-diglucoside(7.34%).The results can provide ideas and references for the utilization of mulberry pomace and the production of anthocyanin products.
