摘 要:试验旨在研究香茅草多糖提取工艺的优化及抗氧化活性分析。以香茅草多糖的提取率为指标,考察超声时间、超声温度、液料比对提取率的影响,依据单因素试验结果,进行响应面试验,得到香茅草多糖的最佳提取工艺,并对香茅草多糖的抗氧化活性进行测定。结果显示,香茅草多糖的最佳提取工艺条件为液料比47 mL/g、超声时间52 min、超声温度44℃,在此条件下多糖的提取率为8.71%。香茅草多糖对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2-联氮-二(3-乙基-苯并噻唑啉-6-磺酸)二铵盐(ABTS+)自由基及羟基自由基的清除率最高分别达到80.02%、82.61%、84.10%,IC_(50)值分别为0.31、0.30、0.27 g/L,香茅草多糖具有良好的抗氧化活性。研究表明,超声辅助热水浸提法适合提取香茅草多糖,提取工艺简便高效,香茅草多糖具有成为抗氧化功能性饲料添加剂的潜力。This study aims to optimize the extraction process of lemongrass polysaccharides and analyze their antioxidant activities.The extraction rate of lemongrass polysaccharides was used as the indicator to investigate the effects of ultrasonic time,ultrasonic temperature and liquid-solid ratio on the extraction rate.Based on the results of the single-factor experiments,response surface methodology was employed to determine the optimal extraction process for lemongrass polysaccharides.The antioxidant activities of lemongrass polysaccharides were evaluated.The results showed that the optimal extraction conditions were a liquid-to-solid ratio of 47 mL/g,ultrasound-assisted extraction time of 52 min,and extraction temperature of 44℃.Under these conditions,the extraction rate of polysaccharides reached 8.71%.The scavenging rates of lemongrass polysaccharides on 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)radicals,2,2-diazo-di(3-ethyl-benzothiazolin-6-sulfonic acid)diamiammonium salt(ABTS+)radicals,and hydroxyl radicals were 80.02%,82.61%,and 84.10%,respectively,with IC_(50) values of 0.31,0.30,and 0.27 g/L,respectively.This indicates that lemongrass polysaccharides possess good antioxidant activity.The study indicates that the ultrasound-assisted extraction method is suitable for extracting lemongrass polysaccharides.The extraction process is simple and efficient,and lemongrass polysaccharides have the potential to be used as antioxidant functional feed additives.
