摘  要:本研究采用Illumina高通量测序技术对丁香杜鹃(Rhododendron farrerae)的叶绿体基因组进行了全面测序与分析,包括基因组特征、密码子偏好性以及简单重复序列(SSR)的鉴定与研究。结果显示,丁香杜鹃的叶绿体基因组大小为149453 bp,不具备典型的四分体结构,基因组序列GC含量为35.65%,基因组总共注释出112个基因,其中74个为蛋白编码基因、4个为rRNA基因、34个为tRNA基因。密码子偏好性分析表明,丁香杜鹃叶绿体基因组更倾向于使用以A和U碱基结尾的密码子,这与其他杜鹃属物种一致,说明自然选择对丁香杜鹃叶绿体基因组中密码子使用的偏好产生了显著影响。此外,共鉴定出270个SSR位点,其中以单核苷酸重复序列最为丰富。本研究为丁香杜鹃的叶绿体基因组提供了重要信息,同时为物种鉴定和遗传结构研究提供了基础数据。In this study,the chloroplast genome in Rhododendron farrerae was comprehensively sequenced and analyzed by using Illumina high-throughput sequencing technology,including the identification and study of genomic characteristics,relative synonymous codon usage,and simple sequence repeats(SSRs).The results showed that the chloroplast genome in Rhododendron farrerae was 149453 bp,without four typical regions,and the GC content of the genome sequence was 35.65%,with a total of 112 annotated genes,including 74 protein-coding genes,4 rRNA genes,and 34 tRNA genes.The analysis of relative synonymous codon usage(RSCU)found that the chloroplast genome in Rhododendron farrerae tends to use codons ending with A and U bases,which is consistent with other Rhododendron species,indicating that natural selection pressure is the main factor affecting relative synonymous codon usage of the chloroplast genome in Rhododendron farrerae.In addition,a total of 270 SSR loci were identified,with mononucleotide repeat sequences being the most abundant.This study provides important information for the chloroplast genome of Rhododendron farrerae,as well as basic data for species identification and genetic structure research.
