摘 要:试验旨在研究间型脉孢菌JY-05211固体发酵豆渣产蛋白酶条件优化及其酶学特性。以豆渣作为发酵主原料,以JY-05211为发酵菌种,采用单因素试验及响应面设计对菌株进行发酵条件优化及酶学性质研究。结果显示,JY-05211固体发酵豆渣产蛋白酶的最优条件为:豆渣50%、麸皮20%、可溶性淀粉30%、酵母浸粉2.8%、Fe2+0.22%、料水比10.0∶7.7、接种量2%、发酵时间72 h,蛋白酶活性为3562 U/g。JY-05211所产蛋白酶的最适反应温度为50℃,最适pH值为7.0,存贮于40~50℃、pH值6~9条件下酶活稳定性较高,Mn2+对酶活促进作用最显著。研究表明,间型脉孢菌JY-05211在以豆渣为主原料培养基中表现出良好的产蛋白酶能力,可生产具有较高蛋白质含量及蛋白酶活性的培养物,实现豆渣的高值化利用。The experiment was aimed at studying the optimization of protease production by solid fermentation of soybean okara with Neurospora intermedia JY-05211 and its enzymatic properties.Using soybean residue as the main fermentation substrate and JY-05211 as the fermentation strain,single-factor experiments and response surface methodology were employed to optimize the fermentation conditions and study the enzymatic properties of the strain.The results showed that the optimal conditions for protease production by JY-05211 solid-state fermentation of soybean residue were as follows:Soybean residue 50%,wheat bran 20%,soluble starch 30%,yeast extract 2.8%,Fe2+0.22%,solid-to-liquid ratio 10.0∶7.7,inoculation amount 2%,and fermentation time 72 h.Under this condition,the protease production activity of Neurosporaintermediawas 3562 U/g.The protease activity preformed optimally at 50℃and pH value 7.0,and the stability was good at 40~50℃and pH value 6~9.Mn2+had the most significant promotive effect on enzyme activity.The study indicates that NeurosporaintermediaJY-05211 demonstrates good protease production ability in a medium primarily composed of soybean residue.It can produce a culture with high protein content and protease activity,thereby achieving the high-value utilization of soybean residue.
