摘 要:为明确沙棘多酚类化合物抑制幽门螺旋杆菌的活性及机理,采用纸片法和肉汤微量稀释法测定沙棘多酚类化合物的最小抑菌浓度,从该类化合物对幽门螺旋杆菌的细胞膜通透性、细胞内容物泄露、膜蛋白和脲酶活力等的影响探究对幽门螺杆菌的抑菌机理。结果表明,表没食子儿茶素和表没食子儿茶素没食子酸酯具有较好的抑菌效果,抑菌圈分别达到17.06、22.59 mm,用最小抑菌浓度处理幽门螺杆菌时抑制率达到50%以上;没食子儿茶素没食子酸酯对幽门螺旋杆菌抑菌效果最好,最小抑菌浓度为156.3μg/mL,用4倍最小抑菌浓度的表没食子儿茶素和表没食子儿茶素没食子酸酯处理幽门螺旋杆菌后,细胞膜通透性分别增加0.68、0.82倍,内容物泄露分别增加0.47、0.48倍,脲酶活力分别降低39%、38%。结果表明,沙棘多酚化合物,特别是表没食子儿茶素和表没食子儿茶素没食子酸酯对幽门螺旋杆菌的生长有抑制作用,具有明显的抑菌活性。To investigate the effect of inhibitory and mechanisms against Helicobacter pylori by polyphenolic catechin compounds from sea buckthorn berries,the minimum inhibitory concentration was determined by the paper disk and broth microdilution.The antibacterial mechanism of polyphenolic catechins from sea buckthorn against H.pylori was explored by examining their impact on cell membrane permeability,leakage of cellular contents,membrane proteins,and urease activity.Results indicated that epigallocatechin gallate and epigallocatechin exhibited good antibacterial ability,with inhibition zones reaching 17.06 and 22.59 mm.Inhibition rate of two compounds of epigallocatechin gallate and epigallocatechin to H.pylori both could reach more than 50%at the minimal inhibit concentration(MIC).The epigallocatechin gallate showed the best antibacterial ability with an MIC of 156.3μg/mL.After treating H.pylori by two compounds of epigallocatechin gallate and epigallocatechin with four times concentration of MIC,the cell membrane permeability increased by 0.68 and 0.82 times,the leakage of cellular contents increased by 0.47 and 0.48 times,and the urease activity decreased by 39%and 38%respectively.The above results demonstrated that sea buckthorn polyphenolic compounds,particularly epigallocatechin,and epigallocatechin gallate,having an inhibitory effect on the growth of Helicobacter pylori and exhibitting significant antibacterial ability.
