摘 要:文章基于目前对寄生性向日葵列当中苯乙醇苷的研究,进一步优化了提取苯乙醇苷的料渣中多糖的工艺。采用单因素试验和响应面法,并建立了回归方程的预测模型。结果表明,最佳工艺条件为浸提温度48℃、超声功率326 W、料液比1:60 g/mL、浸提时间45 min,提取多糖的得率为(2.73±0.03)%。测定列当多糖的纯度和分子质量的结果表明,多糖中不含蛋白质和核酸2种物质,其分子质量为1.528×10^(5) u。抗氧化活性评价结果显示,列当多糖具有清除1,1-二苯基-2-三硝基苯自由基和羟自由基的能力,且清除率与多糖浓度呈正相关。抑菌试验结果表明,列当多糖对大肠杆菌有较弱的抑制作用。该研究为进一步深入研究寄生性向日葵列当的功能活性提供了数据支持。The present study was based on the study of phenylethanol glycosides in parasitic Orobanche cumana Wallr.to further optimize the extraction process of polysaccharide from residue after extraction of phenylethanol glycosides.In this study,singer factor test and response surface methodology were used and a predictive model of regression equation was developed.The experimental results showed that under the optimal process conditions(material-liquid ratio of 1:60 g/mL,extraction temperature of 48℃,ultrasonic power of 326 W,extraction time of 45 min),the yield of polysaccharides was(2.73±0.03)%.The results of determining the purity and molecular weight of Orobanche cumana Wallr.polysaccharide showed that the polysaccharide did not contain both protein and nucleic acid,and its molecular weight was 1.528×10^(5) u.This study of antioxidant activity evaluation showed that Orobanche cumana Wallr.polysaccharide had the ability to scavenge 1,1-diphenyl-2-trinitrophenyl radicals and hydroxyl radicals,and the scavenging rate was directly proportional to the concentration of polysaccharide.The results of the bacteriostatic experiments of Orobanche cumana Wallr.polysaccharide showed that the polysaccharide had a weak inhibitory effect on Escherichia coli.These findings provided data support for further in-depth study of the functional activity of parasitic Orobanche cumana Wallr.
